Objective: The aim of this study was to investigate the expression of CRM1 gene mRNA in multiple myeloma KM3 cell line by using Arsenic trioxide。Methods: Cell proliferation was observed by the Cell Counting Kit-8; Using inverted microscope to observe the cell morphology after the drug and to take a photograph. The cell apoptosis was analyzed by flow cytornetry. To determine the mRNA expressions of CRM1 in KM3 cell line after using different concentrations of As2O3 by RT-PCR;Results: Arsenic trioxide inhibit the proliferation of KM3 cells with obvious dose effects; The apoptosis rates for 10、20、30μmol/L As2O3 after 24 h in KM3 cells were(23.95±0.212)%(24.30±1.698)%(42.15±0.4949)% respectively in KM3 cells. With the control group(5.15±0.212)%,there were significant differences(P<0.05);Compared with control group, the mRNA expression was reduced in As2O3 groups(p<0.05).No significant difference was observed between the every experimental groups.Conclusion: As2O3 can inhibiting KM3 cell proliferation and induce cell apoptosis;CRM1 mRNA expression was decreased.
