| Objective: To observe the therapeutic effects of Jianpi Shugan Jiangzhi Decoction on nonalcoholic fatty liver in mice, clarify its mechanism of action through the indexes such as liver function, hepatic fibrosis etc. and provide a new basis for the application of Jianpi Shugan Jiangzhi Decoction on nonalcoholic fatty liver field in order to better guide clinical application, and develop ideas for further scientific research.Methods: 72 SPF(specific pathogen Free) level Kunming mice were given SPF level diet for 1 weeks, then were randomly divided into 6 groups, namely the normal group, model control group, Kezhi capsule group, Jianpi Shugan Jiangzhi Decoction high, medium and low dose group( n = 12). The normal control group were fed with normal diet, the other groups were fed with high fat feed, and at the same time were given a subcutaneous injection of 10% CCl4 edible oil solution in the lower abdomen(dosage of 0.1m L/10 g, 2 times / week).The normal group and model group were given medical same volume distilled water gavage, The other groups were given the Jianpi Shugan Jiangzhi Decoction liquid or Kezhi capsuleaqueous gavage especially, which were all given 0.2m L/10 g according the body weight for 6 weeks. Fasting 12 h after one treatment, give chloral hydrate solution 4 ml/kg intraperitoneal injection of anesthesia, anesthesia after the success, giving abdominal aorta puncture in blood, the blood samples under the low temperature centrifugal separation and detection of serum alanine aminotransferase(ALT), aspartate aminotransferase(AST), Serum gamma glutamyl transpeptidase(GGT), serum phosphatase(AKP) and liver fibrosis indexes, hyaluronic acid(HA), type IV collagen(CIV), laminin(LN), type â…¢ before the content of collagenâ…¢(PC). When the mice were killed later,the wet liver was weighted, and liver index was calculated. The liver was washed with 0.9% sodium chloridesolution,then the left lobe of the liver was cut at the same position,which were fixed with 4% paraformaldehyde fixation, embedded by paraffin then sliced, stained with HE to observe the pathological and pathology at last.Results:(1) When the model group compared with normal group, the liver index and ALT, AST, GGT, AKP and HA, PC III were markedly increased, there were statistical significant difference(P<0.01).(2) When the treatment group compared with the normal group, there were statistically significant difference in the liver index and ALT, AST, AKP and CIV, LN,HA(P<0.05); when the treatment of low dose and middle dose group compared with normal group there had no statistical significant difference in the lower GGT, LN levels(P> 0.05); In the aspect of reducing PC III, CIV level, there were no statistically significant difference between the treatment dose group and normal group(P>0.05).(3) compared with model group, in addition to the lower levels of LN in high dose group compared with model group, no statistical significance(P > 0.05), the treatment of high, medium and low, shell fat capsule group, the rest of the indicators are significantly reduced or mitigated, was statistically significant(P < 0.05).(4)In the aspect of improving the related indexes of liver: every dose group could reduce the levels of serum ALT, AST, AKP, GGT, HA, PC III, LN, CIV levels, which had significant difference(P<0.05); In the aspect of reducing the level of ALT, HA, GGT, low dose group was better than the Kezhi capsule group, high dose group; In the aspect of reducing the level of AST, middle dose group was superior to the other 3 groups; in the aspect of reducing the level of AKP, middle dose group was best, low dose group was better, high dose group was good, Kezhi capsule group is bad; in the aspect of reducing PC III, CIV level, middle dose group was good but low, high and Kezhi capsule group has certain therapeutic effects.(5)The light microscope: the structure of hepatic lobule was normal, liver cell cords were arranged in order, the radial arrangement exists, peripheral lobular small hepatocellular micro vesicular steatosis, intracellular abundant cytoplasm, nuclear size to normal position centered. Compared with model groups,steatosis degree was greatly improved, and the number of liver cells steatosis was significantly reduced, intracytoplasmic lipid droplets decreased or disappeared with abundant cytoplasm; Partial liver cells compared with normal group in the aspect of cell swelling, cytoplasm withabundant cytoplasm, nuclear size normal, location center, a small amount of fat vacuoles visible. The liver color was bright red, which showed that intrahepatic fat deposition was no obvious. Among the groups in the liver index there were statistically significant difference(P<0.05).Conclusion:(1) Jianpi Shugan Jiangzhi Decoction has a good regulation for ALT, AST, GGT, AKP and HA, CIV, LN, PC â…¢on NAFLD mice which can reduce the serum transaminase activity,enhance the NAFLD mouse liver lesion cell recovery, reduce blood lipid content,and decreased the activity of FFA;(2) Jianpi Shugan Jiangzhi Decoction can reduce the liver appearance and weight of NAFLD mice, so as to improve the degree of fatty degeneration of liver, inhibit liver fibrosis;(3) Jianpi Shugan Jiangzhi Decoction can improve liver function and inhibit liver fibrosis of NAFLD mouse, it associated with the dose of drug.
