Objectives: Research on this topic through animal experiment, to explore the method of steroid-induced necrosis of the femoral head of rabbit model of replication. Acrossing to investigate the effects of total flavonoids of Rhizoma Drynariae on the blood calcium,the phosphorus,ALP,trabecular bone area ratio, rate of empty lacuna and histological pathology of Steroid-induced avascular necrosis of the Femoral Head in rabbite model,in order to clarify the mechanism of preventing or treating Glucocorticoid-induced Avascular Necrosis of Femoral Head treated with total flavonoids of Rhizoma Drynariae and proved a theoretical basis for clinical application of traditional Chinese medicine.Methods: The experiment adopted 24 New Zealand rabbits(female and male in half) as the research object,after the steroid induced necrosis of the femoral head in rabbite model are established,blank group and model group with physiological saline 6ml a day orally once, continued by gawage for 8 weeks, the experiment group is treated by total flavonoids of Rhizoma Drynariae pressing 35mg/kg everyday at a time, a total of 8 weeks. After 8 weeks, the experiment group compared with the model group,the blank group, Acrossing to investigate the effects of total flavonoids of Rhizoma Drynariae on the blood calcium,the phosphorus,ALP,trabecular bone area ratio,rate of empty lacuna and histological pathology of Steroid-induced avascular necrosis of the Femoral Head in rabbite model,To investigate the relationship between ALP,the blood calcium,the phosphorus,and bone cell transformation activity of Steroid-induced avascular necrosis of the Femoral Head treated With Total flavonoids of Rhizoma Drynariae.In order to clarify the related rmechanism of preventing or treating Glucocorticoid-induced Avascular Necrosis of Femoral Head treated with total flavonoids of Rhizoma Drynariae.and observe the changes of trabecular bone area ratio,rate of empty lacuna of Steroid-induced avascular necrosis of the Femoral Head in rabbite model treated with total flavonoids of Rhizoma Drynariae under the light microscope.Methods:Determination of the blood calcium,phosphorus content:the blank group compared with the model group,the differences were statistically significant(P <0.05), the result suggest that a large dose of hormone can make the rabbits of the model group were obviously decreased blood calcium and increased phosphorus significantly, caused the bone cell destruction dissolved formated osteoporosis, and then lead to the occurrence of osteonecrosis of the femoral head and contribute to the formation of new bone or calcification,inhibit bone calcium into the blood metastasis,and prevent the occurrence of osteoporosis. the experiment group compared with the model group, the differences were statistically significant(P <0.05). The model group compared with the blank group, there are no significant difference.It shows that serum Ca and P level basically can be returned to normal level after treated with total flavonoids of Rhizoma Drynariae,at the same time,it proved that Drynaria total flavonoids contribute to the formation of new bone or calcification,inhibit bone calcium into the blood metastasis, and prevent the occurrence of osteoporosis.The activity of intracellular alkaline phosphatase(ALP) was determinated:the model group compared with the blank group, ALP activity of the experiment group decreased,the differences were statistically significant(P <0.05),it was suggested that the bone cell metabolism is reduced, bone resorption increased.Trabecular bone area ratio, rate of empty lacuna:the model group compared with the blank group,the experiment group,trabecular bone area ratio of the model group is at a lower level,there are significant difference(P<0.05);the experiment group compared with the blank group,trabecular bone area ratio of two groups are both higher,there are no significant difference between two groups(P>0.05);the model group compared with the blank group,the experiment group,the rate of empty lacuna of the model group is higher,the differences between two groups were statistically significant(P<0.05);The experiment group compared with the blank group,the rate of empty lacuna of two groups are both lower,there are no significant difference between two groups(P>0.05).Conclusions:Total flavonoids of Rhizoma Drynariae can regulate blood calcium and phosphorus content,contribute to the formation of new bone or calcification, improve the activity of alkaline phosphatase,then promote osteoblast formation action; Acrossing to stimulate the generation of trabecular bone,reduce the rate of empty lacuna,thus reduce bone cell destruction, and eventually realize the control bowel primality avascular necrosis of the femoral head purpose,and provide theoretical basis for clinical treatment of the disease.
