Objective Through the study of acute cardiorenal syndrome laboratory rat models, we observe the influence to the relevant indexs of Qiliqiangxin capsule on function of heart-renal. Evaluating the effect of the treatment of Qiliqiangxin capsule to the acute cardiorenal syndrome laboratory rat models,In order to provide a new theoretical basis for the Traditional Chinese medicine about prevention and treatment of the acute cardiorenal syndrome. Further providing the relevant experimental data for the treatment of Qiliqiangxin capsule clinical application to the acute cardiorenal syndrome.Methods 1.We chose 80 male Sprague Dawley(SD) rats(specific pathogen free,SPF),weight 200±20g. 2.All the rats application of the ultrasonic examination checked the heart function before the surgery. Then they were randomly divided into the sham group(n=8) and the building model group(n=72),with the random number table. Every group intraperitoneal injection of anesthetic after weighed,using the 10% chloral hydrate(3ml/kg),then the building model rats were removed the right kidney.The sham group didn’t remove the right kidney,only had a incision on the back, exposed and sutured after cutting. One week after the right kiney were removed(unilateral nephrectomy,UNX), making the myocardial infarction(MI) rat modle using the method of ligaturing the left coronary artery.The sham group without the ligature,only hang a line on the same position. To prevent infection, both of the groups intraperitoneal injection of penicillin(240 thousand units/kg·d),a total of 3 days. 3.After the successful building model surgery, all living rats were randomly divided into the model group、Qliqiangxin capsule high dose group、Qliqiangxin capsule dose group、Qliqiangxin capsule low dose group.Fasting 6h before lavage the drugs, keep the rats in the fasting state, in order to facilitate the absorption of drugs,every dose group was filled the stomach with the drug liquids maked of 3 ml distilled water(1.2g/kg·d、0.6g/kg·d、0.3g/kg·d),on the second day after the surgery,Once a day, in the morning,on the same time,for four weeks consecutivly.The other groups were filled the stomach with the same volume physiological saline, in the same way.All rats were given regular food and water. 4. All groups ultrasonic examination to check the heart function 24 h after the MI sugery and drug intervention 4 weeks.Measureing the Left ventricular end-systolic diameter(LVDs)、Leftventricular end-diastolic diameter(LVDd)、Cardiac output(CO)、Left ventricular end-systolic volume(LVESV)、 Left ventricular ejection fraction(LVEF)、Left ventricular fractional shortening(LVFS),And the blood test indices included the Brain natriuretic peptide(BNP)、Serum creatinine(Scr)、Urine Urea Nitrogen(BUN)、Neutrophil gelatinase associated lipocalin(NGAL).At last, all rats were killed with the anesthesia and collected the myocardial tissue not the places of infarction, Using the method of Masson trichromatic staining, to observe the myocardial morphology in the level of pathological morphology.Results 1.A total of 42 rats were died during the modeling building process and postoperative irrigation feed. 2.General situation: the food intake of all building model groups rats were reduced, the reactivity and activity were reduced.They resisted weaker when were grasped.The sham group:normal diet and activities. 3.All the results of rats’ heart ultrasound examination: ○1 Before the sugery:Compared to the sham group with the building model group on LVEF、LVFS、CO、LVDs、LVDd and LVESV,there was not any statistically significant differences(P>0.05);○2 4 weeks after drug intervention:Compared to the sham group,the model group and all dose groups were reduced on LVEF、LVFS and CO, increased on LVDs、LVDd and ESV, the difference was statistically significant(P<0.05);○3 compared with the model group, all dose groups were reduced on LVDs、 LVDd and LVESV, and, increased on LVEF 、 LVFS and CO, the difference was statistically significant(P<0.05); ○4 Among all the dose groups, little change, there was no statistically significant differences(P>0.05); 4. T he results of rat myocardial tissue pathology morphology observation:○1 The sham group:myocardial tissue, arranging and the form was normal; ○2 The model group :myocardial tissue disordered arrangement、 hypertrophy,the myocardial was mixed each other between the infarction area and non infarction area.a number of myocardial cells were instead of green collagen fiber, the structure was loose and messy; ○3 Each dose group myocardial cells hypertrophy,less parts of the myocardial tissue were instead of green collagen fiber, Collagen fibers were branching, and crossed arrangement with the myocardial cell,The degree of myocardial fibrosis were lighter than model group, but Significantly greater than the sham group. 5.Serum biochemical index test results:24 hours after the MI:Compared to the sham group,other groups were increased on NGAL, the difference was statistically significant(P<0.05), Illustrating the building model surgery had caused the kidney damage. 4 weeks after the drug intervention:○1 Compared with the sham group,The others were elevated on Scr、NGAL、BUN and BNP, the difference was statistically significant(P<0.05);Illustrating the building model surgery had damaged the kidney and heart;○2 Compared with the model group, high and medium dose group were lowed on Scr 、 NGAL and BUN the difference was statistically significant(P<0.05);but the low dose group had nothing change(P>0.05), there was no statistically significant differences. there was no statistically significant differences between medium dose group and low dose group(P>0.05);all Qiliqiangxin capsule dose groups were low on BNP, the difference was statistically significant(P<0.05), high dose group had a significant differences, the difference was statistically significant(P<0.05); there was no statistically significant differences between high dose group and dose group(P>0.05);there was no statistically significant differences between high dose group and dose group(P>0.05).Conclusion 1.Through the sugery with UNX+MI,we can successfully build the acute CRS rat model. 2.Qiliqiangxin capsule can improve the heart function of the acute CRS rats by increasing the cardiac contraction ability. 3.Qiliqiangxin capsule can improve the renal function of the acute CRS rats by reducing the level of the Scr、NGAL、BUN. 4.Qiliqiangxin capsule can improve the myocardial morphology of the acute CRS rats.
