Objective: To study the effect of EGFR amplification and mutation, PTEN mutation for EGFR and PI3K/AKT signaling pathway gene mRNA expression. To analysis the clinical pathological significance of EGFR and PI3K/AKT signaling pathway mRNA expression, EGFR copy number and mutations of EGFR and PTEN in the patients survival of glioblastoma.Methods:From open reading TCGA website (http://tcga-data.nci.nih.gov) download normalized 160 cases of patients with brain glioblastoma EGFR copy number and the mutations of EGFR and PTEN, the expression of EGFR, PTEN and PI3K/AKT signaling pathway mRNA. And to analyses the effect of EGFR amplification and mutation, PTEN mutation for EGFR and PI3K/AKT signaling pathway gene mRNA expression, and the clinical pathological significance of EGFR and PI3K/AKT signaling pathway mRNA expression, EGFR copy number and mutations of EGFR and PTEN in the patients of glioblastoma by statistic methods.Results:When EGFR amplification, EGFR mRNA low expression was 42 cases (35.0%), overexpression was 40 cases (100%), the difference was statistically significant (P<0.05); PTEN mRNA low expression was 68 cases (56.7%), overexpression was 14 cases (35.0%), the difference was statistically significant (P <0.05); PI3K, AKT and mTOR mRNA expression was not significantly different. When EGFR mutations, EGFR mRNA low expression was 24 cases (20.0%), overexpression was 18 cases (45.0%), the difference was statistically significant (P <0.05); PTEN, PI3K, AKT and mTOR mRNA expression was not significantly different. PTEN mutations for the expression of EGFR, PTEN, PI3K, AKT and mTOR mRNA had no significant effect. The survival rate of patients with overexpression of EGFR mRNA was significantly lower than the low expression (P <0.05). The survival rate of patients with EGFR amplification copy number was significantly lower than normal (P<0.01). The survival rate of EGFR with mutations was significantly lower than those without mutations in the patient (P<0.05). The mutation of PTEN and the expression of PTEN, PI3K, AKT and mTOR mRNA for brain glioblastoma survival rate was not significant effect.Conclusion:Both amplification and mutation of EGFR are applied to the expression of EGFR mRNA in brain glioblastoma, and EGFR amplification, mutation, and the mRNA overexpression all can be used as an important indicator to assess the prognosis of brain glioblastoma. There was no effects of EGFR amplification and mutation on mRNA expression of PTEN, PI3K, AKT and mTOR in glioblastoma.
