| Objective : This experiment by studying the change of vaginal flora and the correlation of high-risk type HPV infection and CIN development, then for regulating vaginal microecological balance, blocking CIN development to provide a certain basis.Methods:1. Collection of dalian from 2014 to 2015 maternal and child health care of outpatient service of department gynaecology treated 130 cases of patients with 20-65 years old, according to the result of HPV detection and cervical biopsy under colposcope grouping pathological diagnosis.2. Using the QIAamp Mini DNA and Blood Mini Kit Kit genomic DNA extraction of vaginal flora.3 by PCR- DGG degeneration gradient gel electrophoresis technology changes in patients with vaginal flora analysis experiment, using the Quantity One cluster analysis software, and the method of using rubber cutting sequencing analysis specificity stripe species category.This experiment specimen under the condition of the research object of informed consent. This experiment mainly by degeneration gradient gel electrophoresis test method that DGGE method to detect CIN patients with vaginal microecological bacteria every period of change. Can be seen from the cluster analysis and PCA analysis, HPV(-) bacteria group and HPV(+) patients As evident in the two groups, suggests that HPV(-) group and HPV(+) bacteria have obvious different, HPV(+) have taken place in the group of patients with bacterial flora imbalance. Steady-state vaginal flora disorder associated with HPV infection.5. Can be seen from the cluster analysis and PCA analysis, HPV(+)bacteria group and CIN patients were dividedinto two groups, significant that HPV(+)group and CIN flora are obviously different.Conclusion:1. The vaginal flora, lactic acid bacteria as dominant fungi.2. Compared with HPV(-) group of HPV(+) group, CIN patients with vaginal bacteria abundance increase.3. Curly lactobacillus, Roy’s lactobacillus, inert lactobacillus is HPV(-)group, the HPV(+) group, CIN group has the advantages of the fungus, belonging to door thick wall; Vaginal bacteria Gardner is HPV(+) group, CIN group of species,belonging to actinomyces.4. Vaginal dysbacteriosis associated with HPV infection. |
