| objective:By evoked potential spinal cord injury in rats after modeling, behavioral science score and ibserved ultrastructural changes in the spinal cord under transmission electron microscope, we valuate Chinese medicine prescription "Ji sui kang" in the treatment of spinal cord injury, nalysis the relationship between evoked potentials and behavioral science score, observed iltrastructural, promote neveral repair and recovery of neurological function effect. Also we esearch the related mechanisms of "Ji sui kang" treatment of spinal cord injury, reduce econdary spinal cord damage and promote recovery of neurological function. It provides an xperimental basis and clinical significance for medicine "Ji sui kang".Methods:There are 41 SD rats in the experiment, of which five rats were continuous evoked potential fter anesthesias to test whether anesthetic affect on the experiment.20 rats were divided into ham operation group, model group, prednisone group, and "Ji sui kang" group with 5 erch. The irst group of rats using modified Allen’s method to establish animal model of spinal cord injury, ham operation group which only bite than laminectomy and not hit the spinal cord. After lodeling the sham operation group and model group were given normal saline, prednisone ;roup and spinal rehabilitation group were given Prednisone solution and "Ji sui kang" decoction ;arage. After each time point in the modeling, CBS function score and evoked potential were onducted on rats lower limbs. The remaining 16 rats were divided into two batches, each batch if 8, by The grouping method above with 2 erch.These rats were received the same molding method and dose regimen. Kill the first batch of 8 rats 2h later after gavaging in the third day and econd batch of 8 rats in the seventh day. Take modeling spinal cord tissue damage zone in the ransmission electron microscope to observe the ultrastructural changes.lesults:The experimental results show that:first, anesthetics on this experiment had little effect on the evoked potential test results. Second, sham group’s CBS scores showed no significant lifference (P> 0.05) at each time point. The values of Prednisone group and "Ji sui kang" group re lower than the model group and showed significant difference (P< 0.05) at each time point, lowever, no significant difference between the two groups of drugs in the intervention group P> 0.05). Third, in evoked potential, Sham group cells evoked potentials declined slightly after nodeling. After modeling 3d returned to normal, and there is no significant difference at all time oints (P> 0.05). After modeling the rest of the groups’evoked potentials were shown to reduce olatility and latency extend, while volatility gradual recovery and latency shorten over time. Each time point, model group amplitude was minimum and latency was longest. There were significant differences (P<0.05) compared with the other groups. There was no significant difference (P> 0.05) between the two groups of drug intervention group at each time point. Finally, observation of spinal cord tissue ultrastructure under electron microscope, we can see that in Sham group spinal cord tissue neuronal cell membrane and nuclear membrane structures were intact, organelles structure intact, morphology clear, no swelling, no significant myelin demyelization phenomenon, no axonal atrophy, and no progressive changes when compared 7d with 3d. Also, model group Under electron microscope, we can see that neuronal membrane rupture, membrane and nuclear membrane structure is not regular, nucleus nuclear chromatin margination, significant swelling of mitochondria and demyelination phenomenon, neuronal degeneration in the 7d, nerve fiber atrophy and less residual. Endoscopic findings of two drug intervention group situation is more similar, and pathological changes nature similar with the model group, but slight degree compared with model group.Conclusion:Chinese medicine prescription "Ji sui kang" can speed up rats’lower limbs motor function recovery after spinal cord injury. And the medicine can inhibit the degeneration and apoptosis of nerve cells in the spinal cord, axonal demyelination and necrosis, atrophy of nerve fibers. Also the medicine Shortening evoked potential return to the normal time, thereby reducing the extent of secondary spinal cord injury, improve the function of spinal cord conduction and promotes the recovery of post-spinal cord function and nerve repair. |
