Object:Traumatic brain injury (TBI) of rats induced by free fall was reproduced to investigate transplantation effect of induced pluripotent stem (iPS) cells on neurological function and long noncoding RNA (LncRNA) expression in rats with TBI.Methods:TBI model of rats was established by using improved Feeney method. cerebral cortex was exposed and subjected to injury with a cushion hit by 50g impact hammer through releasing it vertically from 20cm high place along the guide bar so as to cause the contusion injury in the right parietal lobe of cerebral motor cortex. One week after TBI, iPS were transplanted into the damaged cortexes through local injection. Neurobehavioral scores were evaluated at 3th days,7th days,13th days, 20th days and 27th days after cell transplantation, respectively. Lastly, cerebral cortexes were harvested 3th days after cell transplantation in order to apply for LncRNA microarray analysis that aimed to study LncRNA expression, together with effect on neurological function in TBI rats with iPS transplantation.Results:Obvious neurobehavioral disorders of rats were successfully induced by using 50g x 20cm outside force parameters.the neurobehavioral scores in the cell transplantation group were significantly higher compared with culture medium transplantation group on 3th,7th,20th and 27th days after transplantation, which exhibited a statistical significance. Moreover, LncRNA chip analysis showed that there were multiple upregulated and down-regulated LncRNAs in the code overlap region between LncRNA and mRNA after cell transplantation group compared with medium transplantation group.Conclusions:Stable TBI model of rats were successfully built referring to improved Feeney method. The damage of neurological function was aggravated after iPS transplanted rats. Compared with medium transplantation group, Cell transplantation group exhibited multiple upregulated and down-regulated LncRNAs in the code overlap region between LncRNA and mRNA, which suggested that these LncRNAs may be the key LncRNAs that is related to neurobehavior in iPS transplanted rats following TBI.
