[Objective] To investigate the effects of different doses of fluorine on the tubulin in hippocampus and the microtubule histopathological changes. Meanwhile we try to explore the effects on the protein expression and mRNA expression of Tubulin-a and Tubulin-β to discuss that the efferent on tubulin of different concentration of fluorine.[Methods] In the test,60 healthy (22±2) g male rats were randomLy divided into 4 groups, and named as normal control group (normal diet,distilled water), low dose group (normal diet,25 mg/L NaF-water), middle dose group (normal diet,50 mg/L NaF-water) and high dose group (normal diet,100 mg/L NaF-water), respectively. To record the mouses’s diets and the weight 60 days.The tubulin histopathological changes were observed by Transmission Electron Microscope.The expression levels of tubulin-α and tubulin-β were researched by real-time RT-PCR. In addition, the protein expression of tubulin-α and tubulin-β were studied by immunohistochemical staining and western blot.[Results] (1) Compared with the normal control group,in the low fluoride group,the microtuble are a little less coherent.There are a large number of microtubles.A part of the fracture,well organized,a little microtuble inclined to the main spindle,microtubles section are evenly.Middle dose group microtubules are poor coherence, organizations are more loose, bent and with spindle microtubules are not parallel; high dose group microtubules are poor coherence, serious fracture, loosely organized, arranged density was significantly reduced,the section of microtubules arranged in uniform. The higher fluoride concentration, the greater the impact of microtubule morphology by fluorine.(2) Compared with the normal control group, the mRNA expression of tubulin-α and tubulin-β in low dose group are almost same.The mRNA expression of tubulin-α and tubulin-β are both decrease in middle dose group and high dose group,and those in high dose group decreased significantly (P<0.05).(3) Detected by immunohistochemical staining,in high group,we found that the positive brown product of tubulin-α and tubulin-β are bend and became pieces from thin trips.pyramidal cell layer became thin,loose and irregular.The cells are small and cytoplasm are thin.Compared with the normal control group, the protein expression of tubulin-a and tubulin-P in low dose group are almost same.The protein expression of tubulin-α and tubulin-β are both decrease in middle dose group and high dose group,and those in high dose group decreased significantly (P<0.05).[Conclusion] Fluorine can change tubulin’s forms of hippocampus in mices.Whin the experiment time, low concentration of fluorine which can reduce expression of tubulin protein,while the high concentration of fluorine which can reduce expression of tubulin protein. Different concentrations of fluoride will destroy the mouse cytoskeletal proteins of hippocampus, thereby affect brain development in mice. However, within some time low concentrations of fluoride on the expression of tubulin hippocampus promotion mechanism needs further research.
