Danbeiyifei Recipe’s Influence On The Expression To TGFβ 1mRNA And VEGF In Pulmonary Tissue Of IPF Rats

Objective:The purpose of this subject is to study how DanBeiyifei recipe improve the situation of IPF rats. The mechanism which DanBeiyifei recipe affects IPF will be investigated by observing how TGFβ 1mRNA and VEGF change in the pulmonary tissue of IPF rats.Methods:108 healthy and clean Wistar rats(half male and half female) were raised for one week,and were modeled by bleomycin(BLM).All the 108 rats were randomly divided into six groups(18 rats per group),namely normal control group( normal rats),model control group(rats with normal saline), high dose DanBeiyifei recipe group(rats with 1.2g/ml solution), moderate dose DanBeiyifei recipe group(rats with 0.6g/ml solution), low dose DanBeiyifei recipe group(rats with 0.3g/ml solution),positive medicine control group( rats with 0.5mg/ml prednisone solution).They were all gavaged by 10 ml solution per kilogram. Then, on the 7th day,14 th day,28 th day,6 rats were randomly killed in each team. The lung tissue from the dead rats were observed by the way of Masson dyeing.The expression of VEGF was detected by Immunohistochemistry. The expression of TGFβ 1mRNA was detected by In situ- hybridization.Result:The expression of Danbeiyifei recipe in each dose group and prednisone group TGFβ 1mRNA and VEGF in each time period were lower than in model group(P<0.01); Danbeiyifei recipe high, middle dose group compared with prednisone compared to the control group, no significant difference between the expression of TGFβ 1mRNA and VEGF(P>0.05); Danbeiyifei recipe in low dose group compared with prednisone compared to the control group, the expression of TGFβ 1mRNA and VEGF was significantly higher than that of prednisone group(P<0.05).Conclusion:1.DanBeiyifei recipe can control the expression of TGFβ 1mRNA and VEGF to ease pulmonary fibrosis of rats.2.DanBeiyifei recipe can ease degree of IPF.
