Objective: Perilla is a kind of medical and edible plants with high nutritional and pharmaceutical value. Jilin province is the main Perilla producing areas, The residue was picked after leaves, stems and the like Tori was dumped. In this study, with residual leaf, stem and receptacle for raw materials, less waste that is called basil.The aim of this paper was to research the extraction of the volatile oil from the Perilla waste and the antibacterial and antioxidant activities of the volatile oil. Based on the research results of this paper, more Perilla products can be developed. These products have many characteristics, such as a wide range of raw material, low costs and waste reclamation, and can contribute to the whole economic development in our province.Methods: The first part, taking volatile oil extraction rate as an index to optimize the extraction process of the SFE(supercritical carbon dioxide extraction) research by star point response surface method. The second part was to study the antibacterial activity of the volatile oil. The growth curves of Escherichia coli, Staphylococcus aureus and Candida albicans were analyzed and the antibacterial activity of the volatile oil on them were tested by paper disc method. The third part was to evaluate the antioxidant activity of the volatile oil by the scavenging rate of DPPH, hydroxyl radicals and superoxideanion.Results:(1) The optimum extraction technology conditions of the volatile oil in perilla waste SFE : The extraction pressure, temperature and time are 26 MPa, 56℃ and 77 min, respectively. Under this condition, the extraction rate of volatile oil in Perilla waste can be 6.718%.(2) The crude extract of the perilla waste extracted by SFE had no obvious inhibitory action on Escherichia coli, Staphylococcus aureus and Candida albicans, while it showed potent antioxidative effect on DPPH, superoxide and hydroxyl radical(the IC50 of their scavenging rate were 9.645, 2.897 and 4.509mg/m L respectively). The refining volatile oil of the crude extract had shown more antibacterial activity and more antioxidant activity(the IC50 of their scavenging rate were 0.196, 0.058 and 0.115mg/m L respectively).Conclusions:(1) Compared with stream distillation, more volatile oil was extracted from perilla waste by SFE. So the optimum extraction conditions of the volatile oil in perilla waste were optimized by SFE combining with star point response surface method. This method is efficient, simple, accurate, fast, and high extraction efficiency.(2) The refining volatile oil had stronger inhibiting effect on Escherichia coli, Staphylococcus aureus and Candida albicans than the crude extract of the perilla waste by SFE.(3) The results of the scavenging rate of DPPH, superoxide and hydroxyl radical have shown that crude extract of perilla waste by SFE had antioxidant activity, and the antioxidant activity would be much more stronger after the crude extract were refined. |
