| Objectives To investigate the distribution of genotype and allele of the two loci rs10118757 and rs7027989 on c(MTAP) gene, and to investigate the relationship between the two single nucleotide polymorphisms(SNP) and ischemic stroke(IS) in Chinese Hans. It would provide important reference for early diagnosis and treatment of IS.Methods Case control study method was applied. 297 patients with large vessel ischemic stroke were enrolled as case group. 313 healthy subject were enrolled as control group. All of the subjects were Han ethnic. Age and gender were matched between case group and control group. General information of the subjects was recorded. DNA samples were prepared from venous blood. Two SNP loci rs10118757 and rs7027989 in MT AP gene were selected to be candidate research loci. Genotyping were performed by using the Genome Lab SNPstream genotyping platform. The distribution of the genotypic frequencies and allelic frequencies were described. Further analysis were performed to deduce if the two SNPs were associated with the prevalence of IS.Results In total subjects, the frequency of minor allele G at rs10118757 in case group was higher than which in control group, but there w as no statistically significant difference(P>0.05). After stratified by gender, the difference remain no significant(P>0.05). In total subjects, the frequency of minor allele G at rs7027989 in case group was higher than which in control group, but there was no statistically significant difference( P>0.05). After stratified by gender, the difference remain no significant(P>0.05). We found age, body mass index, smoking history, hypertension history, diabetes history and serum LDL-C abnormal were independent risk factor of IS by multivariate Logistic analys is(P<0.05).Conclusions MTAP gene polymorphisms were not associated with the prevalence of IS in Chinese Hans. |
