Objectives: To evaluate effect of N-acetylcysteinein on pulmonary interstitial fibrosis,we investigated pathological change of lung and genetic and protein expression of Smads(3,4 and 7)gene in bleomycin-induced pulmonary interstitial fibrosis in rats which were treated with N-acetylcysteinein.Methods: sixty female Wistar rats were randomly divided into four groups: a bleomycin-induced-fibrosis group(Group a), a bleomycin-induced-fibrosis group treated with NAC(Group b),a bleomycin-induced-fibrosis group treated with Hydrocortisone Sodium Succinate(Group c)and the control group(Group d), which was treated with saline. On the 7th and 14 th and 28 th day, 5 rats of every group were randomly executed, the pulmonary pathological change under HE staining and the change of m RNA and protein expression of Smad3 and Smad4 and Smad7 were observed in lung,by immunnohistochemical method. Reverse transcription polymerase chain reaction(RT-PCR) was used to detect the expression of Smad3 and Smad4 and Smad7 m RNA in the left lung.Results:Alveolitis and pulmonary fibrosis in rats different periods : group d had no significant alveolitis and pulmonary fibrosis; group a of alveolitis and pulmonary fibrosis heaviest,group b followed(P<0.05), group c alveolitis and pulmonary fibrosis compared with group b further reduce(P<0.05). m RNA and protein expression in each group Smads: group a of Smad3/4 m RNA and protein expression of the strongest,group b followed(P<0.05), Group c further reduced compared with group b(P <0.05),group d the lowest group(P<0.05).group c of Smad7 m RNA and protein expression of the strongest,group b followed(P<0.05), Group a further reduced compared with group b(P <0.05),group d the lowest group(P <0.05).Conclusions:The expression of GRP 78 and HSP 27 in NSCLC tissues is associated with metastasis of the cancer, while there is no significant correlation between their expression levels and metastasis degrees.
