In this study,Rosa multiflora Thub as the research object,the extraction and purification methods,and the antioxidant and antibacterial activities in vitro of Total Saponins from Rosa multiflora Thub had been researched.This is a relatively comprehensive and systematic study.The main experimental results in this paper were listed as follows,respectively.(1)The extraction technology were investigated systematically,the comparison of the thermo-refluxing method and advanced technology with respect to microwave-assisted,ultrasonic-assisted,ultrasonic-microwave assisted extraction were conducted in this article.the optimized technology parameters were studied,respectively.The optimum extraction parameters and extraction method of thermo-refluxing extraction were:reflux temperature of 75℃,solid-liquid ratio of1:25 g/m L,extraction time of 3 h min,ethanol concentration of 70%;The optimum extraction parameters and extraction method of microwave-assisted extraction were:microwave power of 450 W,solid-liquid ratio of 1:11 g/m L,extraction time of12 min,ethanol concentration of 70%;The optimum extraction parameters and extraction method of ultrasonic-assisted extraction were:ultrasonic power of 270 W,solid-liquid ratio of 1:25 g/m L,extraction time of 37 min,ethanol concentration of85%;The optimum extraction parameters and extraction method of ultrasonicmicrowave assisted extraction(UMAE) were70% ethanol as solvent and 1:8 g/m L solid to liquid ratio for 3 min at the power of microwave of 100 W,which was higher than that by microwave-assisted extraction(MAE),thermo-refluxing extraction(HRE)and ultrasonic-assisted extraction(UAE).This optimized process is simple,stable and efficient.(2) Purification of the total saponins from Rosa multiflora Thunb by macroporous resin.The static adsorption rate and the desorption rate as the index to investigate purification effect of total saponins from Rosa multiflora thunb by macroporous resin,The results showed that D101 resin showed the best performance.The purification conditions were specified as the following: based on adsorption time6 h of sample solution with an initial concentration of 4.0 mg/m L at p H 6.0,followed by desorption using 70%(v/v) ethanol at p H 7.0 with a ratio of sample solution volume to resin weight of 12:1(m L/g) and eluent flow rate was 3BV/h.(3)The antioxidant activities of the purified Saponins were evaluated in vitroby scavenging capability of DPPH free radical,superoxide anion free radical and hydroxyl free radical and reducing power.The results showed that The purified Saponins from Rosa multiflora Thub had stronger reducing power than VC,the compounds also had the ability to scavenge ·OH,DPPH· and O2-·.(4)Five pathogenic bacteria(Bacillus subtilis,Escherichia coli,Staphylococcus aureus,Proteus and white slime molds) were selected to in bacteriostasis experiments in vitro.The antibacterial activity of purified Saponins from Rosa multiflora Thub on microorganisms and the effect of p H value,temperature,ultraviolet irradiation and culture time on its antibacterial activities were discussed by the method of disk diffusion and standard tube dilution.The results showed that the purified Saponins had the strong inhibition on Escherichia coli with a MIC of 0.121 mg/m L.It had significant inhibition of Bacillus subtilis,Staphylococcus aureus,Proteus and white slime molds,for all MIC were 0.37 mg/m L.The antibacterial activity of Saponins from Rosa multiflora Thub was stable under heating and the Saponins had the best antibacterial effects at p H 5~6,with UV irradiation 20~60 min and culture time18h~36.
