| In this paper, the moisture, water extract, alcohol extract, ether extract,crude fat, crude protein, ash, inorganic elements and amino acids of five velvet antlers were measured. The 8 kinds of inorganic elements in five velvet antlers were determined by an atomic absorption spectrophotometer and 17 kinds of amino acids were determined by spectrophotometry after being separated by an amino acid analyzer. The similarities and differences of different antler composition were revealed by component analysis. The results would provide the theory basis for quality control and pharmacological research of velvet antler.By comparing three kinds preparation methods of polypeptide, polypeptide extracted with p H12 Na2HPO4-Na OH buffer solution protein content of 92.3%, there were four bands and the molecular weight between the 2-30 k Da. Separated by DEAE ion exchange chromatography, Sephdex G-75 filtration chromatography, high performance liquid chromatography to obtain VAP-1. HPLC spectra had a single peak. The results of Ticine-SDS-PAGE showed a single band between the 6.5-14.3 k Da, showed that VAP- 1 was single component.Study on New Zealand red deer velvet antler polypeptides on phagocytosis of macrophages in mice. The chicken red blood cells and macrophages mixed in the test tube to simulate in vivo environment that determined to be swallowed chicken erythrocytes. Compared with the control group, each experimental group phagocytic percentage, phagocytic index were highly significant difference(P<0.01), There were significant differences between the experimental groups(P <0.05). Polypeptide can significantly improved phagocytosis ability of peritoneal macrophage in mice to enhance its non-specific immunity. Phagocytic percentage and phagocytic index increased with peptides dose increasing.Provided scientific basis for adjusting the immune effect of pilose antler polypeptides.Study on New Zealand red deer pilose antler polypeptides on proliferative effect of splenic lymphocytes of mice. The experiment was divided into control group, PAP group, Con A+PAP group, LPS+ PAP group. Effect of peptides on the immune function in mice used the MTT assay. The results showed that peptides can promote effect on mice spleen lymphocyte proliferation in vitro and collaborative Con A, LPS promote T, B lymphocyte proliferation. PAP could improve the immune function.Fatigue model prepared by swimming test. This article determined the effects of PAP on swimming time, liver glycogen, muscle glycogen, serum urea nitrogen, lactic acid, LDH, MDA, SOD, GSH-Px. The results show that pilose antler polypeptides can increased the content of liver glycogen and muscle glycogen, significantly reduced the metabolic product of LA, BUN, MDA levels, increased LDH, SOD, GSH-Px activities, accelerated the clearance of the free radical, enhanced mice antioxidant capacity, improved exercise ability of mice, played antifatigue effect. |
