Objective:Immunocytochemistry method is applied to detect human papillomavirus infection in cervical exfoliated cells, while detecting the expression of autophagy Beclin1 gene level and studying its correlation. In order to provide a new way of Cervical cancer HPV screening for economic less developed areas, because it is simple, low-cost, at the same time, add a new effective auxiliary index.Methods:1500 outpatients(21-65 years old) without the history of Cervical Diseases were randomly selected in the department of gynecology and obstetrics in Shanxi Academy of Medical Sciences, Shanxi Da Yi Hospital during May 2014 to November 2014. We had two cell specimens from cervical squamous column junction and cervical tube at the same time.A sample for Liquid-Based Cytology Test, another samples for hybrid capture of the second generation(HC II) method to detect HPV infection. Using recycle liquid thin-layer cytology test(LCT) after samples of residual liquid, for immunocytochemistry(ICC) method to detect human papilloma virus(HPV) protein expression; Detecting the protein expression of Beclin 1 levels at the same time, All cytologic examination result is Atypical squamous cell of undetermined significance(ASCUS) and Higher level lesions.We had Colposcopy biopsy. Detailed records or more groups of participants’ age, health status, liquid thin-layer cytology(LCT), HPV detection, histopathological examination, Beclin 1 the expression level of test results.Various test results above will check after the entry of SPSS software for statistical analysis and comparison, verification of HPV testing new ways and Beclin 1protein expression a auxiliary index of the clinical application value.Results:1.In 1500 patients, the result of LCT was 210 abnormal(14.0%); the positive HPV infection was 190 cases with the method of Hybrid capture of the second generation(HC II)(12.7%). the positive HPV infection was 140 cases with the method of HC II in abnormal result of LCT(12.7%). Using ICC method is 95 cases(45.2%); Two kinds of test method of statistical processing(c2=85.95,P>0.05); Sensitivity:47.4%; Exdegree:75.0%. 2.Normal cervical epithelium, cervical intraepithelial neoplasia(CIN) I, cervical intraepithelial neoplasia(CIN) II-III of the cervix cells exfoliated Beclinl protein positive expression rate of 56.0%(56/100), 49.1%(28/57), 20.0%(3/15)。Two independent sample test shows that there was no significant difference of Beclinl expression in CIN Iand normal cervix cervical exfoliated cells(P<0.05);Respectively compared with CINII-III, then Beclinl expression level difference significant(P< 0.05). 3.The correlation between HPV infection and Beclin1 protein expression, chi-square test showed that c2= 20.198, P < 0.05, and the experimental results show that the overall chi-square have statistically significant, and with the increase of the HPV virus load value, Beclin 1 protein expression is on the decline.Conclusions:1.Immunocytochemistry was used to detect cervical exfoliated cells of human papillomavirus(HPV) protein expression, It can be used as a selection of HPV detection method in cervical cancer screening in rural areas, nearly 50.0% sensitivity of the screening and speciality rate by 75.0%; And we use a test samples to get wo information(cytological examination and HPV infectiont), for the economy of underdeveloped regions in cervical cancer screening has certain application value; 2.Beclinl Protein expression in cervical exfoliated cells were down-regulation in cervieal invasive careinoma,Prompting Beclinl loss associated with the occurrence of cervical cancer, It can be used as an effective auxiliary index in cervical cancer screening; 3.With the increase of the degree of HPV infection Beclin1 protein expression is on the decline, suggesting reducing function of autophagy may be one of the mechanisms of HPV infection causes cervical cancer.
