The Effects Of Fecal Microbiota Transplantation On Expression Of Some Cytokines In C57BL/6 Mice With DSS-induced Colitis

Objective: To observe the effects of fecal microbiota transplantation(FMT) on mice ulcerative colitis induced by DSS,and to test the expression of IL-1β、TNF-α、IL-4、 IL-10 cytokins in mice blood and colonic mucosa.Methods: Forty C57BL/6 male mice were randomly divided into two groups with a ratio of 1:3, group A(blank control group, n=10) and group S(study group, n=30). The mice in group A were free to drink sterile distilled water from the first day, and from the 11 th day were received sterile saline enema(0.5ml/time, 2 times/day, 6 times in total). The mice in group S drinked 2% DSS solution for 7 days, and then were divided into subgroup B(model control, with enema of sterile saline), subgroup C(enema of Bifico 2548 mg/kg), subgroup D(enema of fecal filtrate, 15.197g/kg*d, 0.5ml/time, 2 times/day, 6 times in total).The general situation, body weights, and survival rate were recorded. The disease activity index(DAI) and the colonic histopathology were evaluated. The concentrations of serum and colonic mucosal IL-1β、TNF-α、IL-4、IL-10 were detected by ELISA method.Result: 1.After modeling was completed(at the 8th day), the DAI score in group A was 0.00±0.00,group B was 1.97±0.81,group C was 2.14±0.63 and group D 2.27±0.52. Compared with the DAI in group A(blank control group), the DAI in B(model control group), C(Bifico group), or D(FMT group) was increased(P=0.000<0.05).At the 16 th day after enema therapy, the DAI score in group A was 0.17±0.28,group B was 1.57±0.93, group C was 0.54±0.43, and in group D was 0.46±0.43. And the histopathological scores(HS) in group A was 0.33±0.52,group B was 1.63±0.74,group C was 0.67±0.52,and in group D was 0.57±0.53. Compared with the DAI and HS in group B, the DAI or HS in group C and group D were significantly reduced respectively(p<0.05), but there was no significant difference between group C and D(P>0.05).2. The serum TNF-α level in group A was 98.52±6.75(pg/ml), group B was138.28±16.82(pg/ml),group C was 118.96±10.41(pg/ml),and in group D was 98.96± 7.45(pg/ml); the serum IL-1β in group A was 40.8±3.96(pg/ml), group B was 66.8±8.49(pg/ml), group C was 49.87±5.78(pg/ml),and group D was 54.48±5.91(pg/ml). The serum IL-4 in group A was 76.1±4.33(pg/ml),group B was 31.6±5.03(pg/ml),group C was 53.9±5.77(pg/ml),and group D was 48.87±5.96(pg/ml);the serum IL-10 level in group A was 165.28±22.03(pg/ml),group B was 96.27±25.24(pg/ml),group C was 149.15±25.45(pg/ml), and in group D was 170.95±16.99(pg/ml). Compared with those in group B, TNF-α、IL-1β’s concentration in group C and group D were decreased(p<0.05), however, the IL-4 and IL-10 level were increased respectively(p<0.05). TNF-α level in group D was lower than in group C(p<0.05), while the concentration of IL-1β、IL-4 and IL-10 between the two groups were not statistically significant.3.The colonic TNF-α level in group A was 1595.75±243.26(pg/ml),group B was 2322.50±278.29(pg/ml),group C was 1836.14±454.61(pg/ml),and group D was 1612.57±337.98(pg/ml); the mucosa IL-1β level in group A was 877.27±167.63(pg/ml),group B was 1304.14±284.01(pg/ml),group C was 898.40±211.22(pg/ml),and group D was 901.5±212.19(pg/ml);the mucosa level of IL-4 in group A was 734.85±128.44(pg/ml),group B was 501.43±124.79(pg/ml),group C was 277.8±98.73(pg/ml),and group D was 377.14±112.07(pg/ml);the intestinal mucosa level of IL-10 in group A was 2373.87±265.66(pg/ml),group B was 1232.00±278.60(pg/ml),group C was 1343.43±235.64(pg/ml),and group D was 1184.57±313.62(pg/ml). Compared with level of mucosal cytokines in group B, group C and group D were reduced respectively(p<0.05),however, no obvious difference between those in group C and group D were seen(p>0.05). The IL-4 and IL-10 expression in group B、C、D had no statistical significance respectively(p>0.05).Conclusion: The C57BL/6 mice ulcerative colitis induced successfully by drinking 2% DSS solution was alleviated by enema of fecal filtrate. Its role may be inhibite the expression of TNF-α and IL-1β.
