Objective To elucidate the roles and the effects of continued light, melatonin antagonists and calmodulin antagonist on the production of bipedal rat scoliosis model and the change of the Cobb angle and the calmodulin levels in the rats’ platelet, which may get a relation between the melatonin,calmodulin and adolescent idiopathic scoliosis,and provide an important animal models and the experimental basis for further study of their pathogenesis. Methods 32 one-month-old female and weaning rats were made into a bipedal rats,whichwas being fed in high cage, prompting its bipedal walking, and were randomly divided into four groups, A group were given luzindole + continuous light,and group B simple to inject luzindole, group C given luzindole + tamoxifen, D group were given the same volume normal sali. The doses of luzindole are 0.2mg / kgand the doses of tamoxifen are 10 mg / d. Taken the X-ray filmsin 8th week and 16 th week and comparisons the different about the success rates and the severity in each group.The thrombocyte is dissociated by centrifugal separation,and the calmodulin from thrombocyte is mensurated by the method of ELISA. Results ① 8th week, the groups of A, B have occurred scoliosis, and the presents were 75% and 12.5%. the degree of scoliosis ranges were 10.8 °-16.8 °, the mean was 12.4 ° in A group and the degree and the mean was 19.4 °± 0° in B.There was a statistically significant difference in the groups A and B(P <0.05).The groups of C,D have neither developed scolisis, scoliosis is unpresent in 100%.②16th weeks, the present was 57% and the degree of scoliosis ranges were 10.1 °-17.9 °, the mean was 14.3 °in A group. the present was 62.5%, and the degree rangs were 18 °-30.3 °, the mean was 25.2 ° ± 4.92 °.The groups of C,D have neither developed scolisis, scoliosis is unpresent in 100%.There were a statistically significant difference in B、Cand B、D(P <0.05) ③There was a significant difference in the calmodulino of Band D,which are from the thrombocyte of aorta blood(P <0.05) ④There was a significant difference in the calmodulino of Band A,which are from the thrombocyte of aorta blood(P <0.05) ⑤There was not a significant difference in the calmodulino of Band C,which are from the thrombocyte of aorta blood(P> 0.05) Conclusion ①The scoliosis models of bipedal rats were found successfully,and confirmed that the generation of scoliosis was concerned with the function of the melatonin. ②The contiuned light can increase the occurrence of scoliosis, but this is reversible ③The lighting conditions Lighting conditions can not increase the incidence of scoliosis and scoliosis deformity finally ④Calmodulin antagonists can effectively inhibit the natural progression of scoliosis, which relate to the function of the calmodulin ⑤The level of calmodulin from thrombocyte cann’t be changed by calmodulin antagonists ⑥The level of calmodulin from thrombocyte had no relation with the severity of idiopathic scoliosis ⑦ The level of calmodulin from thrombocyte can be reduced by melatoninantagonists ⑧ The lighting can reduced the level of calmodulin significantly...
