| Objective: To investigate the pharmacodynamics effects of anti-inflammation, analgesia, choleretic, anti-pyretic of Danshiliuhao Decoction.And to investigate the therapeutical effect of Danshi Liuhao decoction of Guinea Pigs cholecystitis induced by lincomycin hydrochloride. And explore the possible mechanisms of action. Methods: 1. The antipyretic activity was observed in rats with hig temperature induced bysubcutaneous injections 2, 4-dinitrophenol and dry yeast suspension. The analgesiceffects were evaluated with tail flick test, and methanal test of mice.Theanti-inflammatory effects were conducted by xylene-induced ear edema in mice,capillary permeabilityin abdominal cavity of mice. The movement of carbon powder inintestines and the weight of cholecyst in mice were observed to assess the cholagogic. 2. Experiment Guinea Pigs cholecystitis induced by lincomycin hydrochloride wasestablished. The Pigs were treated and then observed the effect on their weight,liverweight, gallbladder weight,gallbladder volume, gallbladder and liver pathologicaltissue,white blood cells and liver function. 3. the content of TNF-α,COX-2 in the serum and bile were measured with enzyme-linked immunosorbent assay(ELISA).The m RNA levels of TNF-α,COX-2,NF-κB inthe gallbladder and liver were detected by real time fluorescent(quantitative(RT-PCR),and the effect of the liver on the NF-κB expression were detected by Western Blotting. Results: 1. The decoction(10,20g/kg) showed antipyretic activity in rats with high temperatureinducedby 2, 4-dinitrophenol(P<0.05, P<0.01). The decoction(10,20g/kg) showedcertain activity in rats with high temperature induced by dry yeast suspension(P<0.05, P<0.01).To varying degree, the doges(10, 20, 40 g/kg) could reduce thesecond phase responses of mice methanal test, The doges(40 g/kg)could increasemouse threshold(P<0.05, P<0.01). The dose(10, 20,40g/kg) decoction could inhibit theinflammatory response induced by xylene, the inhibition ratio is 17%, 32%, 38%(P<0.05, P<0.01). It can reduce the capillary Permeability, the inhibition ratio is 19%,23%, 32%(P<0.05, P<0.01). The dose(20,40g/kg) decoction also could significantlyincrease the biliary secretion, also had promote action on small intestinal motilitydisturbance in mice(P<0.05, P<0.01), the dose(10,20,40g/kg) could stimulate the flowof bile in mice(P<0.01). 2. Compared with the model group, the Danshi Liuhao decoction could gain weight(P<0.05, P<0.01). The dosage could obviously relieve the liver index and gallbladderindex, promote biliation and ease cholestasis(P<0.05, P<0.01). It could reduce themumber of white blood cells and neutrophil(P<0.05, P<0.01),evidently decrease theserum activity of AST,ALT,ALP,GGT,TBIL,TBA(P<0.05, P<0.01) and protectthe liver with a dose-dependent. It also had obviously slighter inflammation of liver andgallbladder. 3. Compared with model group, the treatment groups content of TNF-α,COX-2 in theserum and bile reduced(P<0.05, P<0.01), the expression of TNF-α,COX-2,NF-κBm RNA in the the gallbladder and liver of the guinea pigs appearentlydecreased(P<0.05, P<0.01),the expression of NF-κB protein also obviouslyreduced(P<0.05, P<0.01). Conclusion: 1. The Danshiliuhao decoction has significantanly anti-pyretic, analgesic,anti-inflammatory and choleretic. 2. The Danshi Liuhao decoction has relieved the inflammation; it also has the effect inincreasing bile secretion and protecting liver, treatment function to the Guinea Pigscholecystitis 3. The danshiliuhao decoction on cholecystitis in guinea pigs might be related to thedecreased NF-κB expressions and thus inhibited the expression levels of TNF-α,COX-2. |
