Study On The Preparation Process And Hypnotic Effect Of Zaowei Granules

Zaowei granules was composed of Fructus schisandra chinensis and Semen ziziphi spinosae. The extraction process of zaowei granules was designed on their sedative and hypnotic ingredients. The optimization experiments were designed from extracts and monomer component of schisandra lignans, jujuboside and semen flavonoids, and the ingredients of them were also as the measured indicators. UV spectrophotometry was used to detect the total content of schisandra lignans, semen saponin, and semen spinosin, meanwhile, the HPLC-UV was used to determine the monomer of lignans, jujuboside A, jujuboside B and semen spinosin content, and the extraction process of schisandra lignans, Jujubosides and semen flavonoids was preferred by the the method of orthogonal test and central composite design-response surface. The optimum extraction technology of Schisandra chinensis was that 15-fold amounts of ethanol(92%) ultrasonic extracted for 2 times and 40 min per time; the optimum extraction technology of Ziziphi spinosae semen was that 20-fold amounts of ethanol(70%) reflux extracted for 2 times and 2 h per time. The zaowei granules was combined the extract of fructus schisandra chinensis and semen ziziphi spinosae, added appropriate amount of excipients, and pelletized. The hypnotic effect was evaluated by its synergy effect of zaowei granules and pentobarbital sodium on sleeping in mice. The results indicated that the sleeping time was prolonged obviously and the sleeping latency was shortened by zaowei granules. In this paper, the optimized extraction technology of Fructus schisandra chinensis and Semen ziziphi spinosae, and the preparation of zaowei granules could provide theoretical basis for comprehensive development and utilization on the sedative efficacy by combination of Schisandra chinensis fructus and Ziziphi spinosae semen.
