Objective:To explore enzyme linked immunosorbent assay( ELISA) to detect the concentrated cerebrospinal fluid 6000 early secretory antigenic target(ESAT-6) of mycobacterium tuberculosis value to the diagnosis of tuberculous meningitis.Methods:1. 106 copies of cerebrospinal fluid were gotten using lumbar spinal puncture. Among them 49 copies from the TBM patients and 57 copies from the non-TBM patients.2.The cerebrospinal fluid were concentrated using Amicon Ultra-4 Ultrafiltration centrifugation.3. The ESAT-6 antigen in cerebrospinal fluid was detected using Indirect ELISA assay. Results:1. Using the Amicon Ultra-4 Ultrafiltration centrifuge tube concentrate cerebrospinal fluid,it could concentrate the cerebrospinal fluid samples 30 ~ 60 times in 40 ~ 50 minutes.2. The positive cut off of the ESAT-6 antigen in this study is 1.5625ng/m L.3. The detection sensitivity of ESAT-6 antigen of the concentrated cerebrospinal fluid was significantly higher than the primary cerebrospinal fluid(87.50% compare to 67.35%, P<0.05),and no statistical difference between the detection specificity of ESAT-6 antigen in concentrated cerebrospinal fluid and primary cerebrospinal fluid(92.16% compare to 96.49%,P>0.05). Conclusions:The detection rate of ESAT-6 antigen can be markedly increased by using centrifugal ultrafiltration technology to concentrate cerebrospinal fluid via ELISA detection, and the diagnosis rate of tuberculous meningitis can be increased. Therefore,the concentration of ESAT-6 antigen in cerebrospinal fluid can be improved by this centrifugal ultrafiltration technology,and it is easy operated together with the Indirect ELISA.The method of this study is helpful for clinical application, and it can also improve the diagnostic rate of tuberculous meningitis. |
