Establishment Of Cardiomyocyte Hypertrophy Induced By Angiotensin â…¡ In H9c2 Myocardial Cell Line And Changes In Ubiquitinated Desmin Levels

Objective:Establishment the model of angiotensin II inducing cardiomyocyte hypertrophy in H9c2 myocardial cell, and observing the changes of junction protein ubiquitination level.Methods:Use angiotensin II to induce cardiomyocyte hypertrophy in H9c2 myocardial cell;Cultured in high glucose DMEM medium containing 10% FBS for 48 h, then isolated and subcultured. They were divided into three Ang II concentration groups( 10-8, 10-7, 10-6mol/L) that were dealt with in different hours( 0, 6, 12, 24, 48,72h),and there is also a control group; measured the total protein content use BCA,measured the number of cardiomyocytes in the same area, and calculate the relative surface area of myocardial cells.Observed ubiquitination junction protein levels. Isolated and subcultured H9c2 myocardial cell, divided into four Ang II concentration groups( 0, 10-8, 10-7,10-6mol/L) and were dealt with in different hours( 0, 6, 12, 24, 48, 72h); The expression of ubiquitinated desmin determined by Western blot and immunohistoc-hemistry.The statistical data is represented by x±s, analysis used SPSS14.0 software,comparing the data of two group adopted two independent sample t-test.Results:H9c2 cardiomyocyte hypertrophy was time- and concentration-dependent induced by Ang II,where total cellular protein content,relative cell surface area,the number of cells and the expression of ubiquitinated desmin were significantly increased compared with the control group(P <0.05), which were most significant in10-7mol/L Ang II for 48 h.Conclusions:Cardiomyocyte hypertrophy model was successfully established,where H9c2 cardiomyocyte hypertrophy was induced by 10-7mol/L Ang II in 48 h,and ubiquitinateddesmin were significantly increased.
