| Objective:To investigate the effect of transection of the cervical sympathetic trunk(TCST)on humoral immunity and complement C3 in diabetic rats.Methods:Ninety male Sprague-Dawley rats,aged 3-4 months,weighing 280-300 g,were randomly divided into 3 groups(n=30 each):control group(group C),diabetes mellitus group(group DM), diabetes mellitus+ transection of the cervical sympathetic trunk group(group TCST). Diabetes mellitus was induced by intraperitoneal injection of1% streptozocin(STZ) 60 mg/kg(successful induction of diabetes was defined as blood glucose ≥16.7 mmol/L) in group DM and TCST, while the equal volume citrate buffer was injected intraperitoneally in group C. In group TCST right transection of cervical sympathetic trunk(TCST)was performed after 1 week of the model of diabetes successfully induced, while Group C and group DM only suffered from the exposure and separation of the right cervical sympathetic nerve trunk but not cut. Six rats were chosen randomly from each group before TCST(T0),and on 1st(T1),3rd(T2), 5th(T3),7th(T4) week after TCST, the blood glucose was recorded, the concentration of serum immunoglobin A,G, M(Ig A, Ig G, Ig M) and the complement C3 were detected by enzyme linked immunosorbent assay(El ISA),the body weight and weight of spleen were measure for spleen index.Results:(1)Compared with group C, blood glucose and the concentration of serum Ig A,complement C3 were increased, the concentration of serum Ig G, Ig M were decreased at T0-T4(P<0.05) and spleen index was decreased at T2-T4 in DM and TCST groups.(2)Compared with group DM,the concentration of blood glucose was decreased at T1-T4 in group TCST; the concentration of serum Ig A, complement C3 were decreased,the concentration of serum Ig G, Ig M and spleen index was increased at T2-T4 in group TCST(P<0.05).Conclusion:The humoral immunity was decreased in diabetic rats, transection of the cervical sympathetic trunk(TCST) can decrease blood glucose,serum Ig A and complement C3 concentration, increase serum Ig G, Ig M concentration and spleen index, which can improve the humoral immunity and complement C3 in diabetic rats. |
