Taxol is a tricyclic two terpene compound isolated from Taxus plants, its unique anticancermechanism, have special curative effect to many kinds of cancer.Taxus madia in the natural taxus is a hybrid variety,its male parent is the Taxus baccata,The mother is the Taxus cuspidata.Because of its own in the species ecological natural advantages, and through the development of a large number of artificial cultivation,at present,Taxus madia has become one of the preferred Taxus species for the extraction of taxol.Now a large number of Taxus madia have been introducted in china,But before the most of raw material extracted taxol came from the various natural Taxus bark, Then the extraction of taxol has its unique advantages in from the branches and leaves of Taxus madia.This article is for the extraction process optimize of taxol from Taxus madia’s branches planted in Jiangxi province Jiujiang De’an County,And the taxol content was determined by hplc.This experiment is combined with TLC and HPLC to research The taxol in the branches and leaves of Taxus madia.This paper is divided into five chapters:The first chapter are reviewed mainly to study the situation of taxol and Taxus chinensis、the research of paclitaxel detection analysis method and extraction methods;The second chapter is the qualitative experiment to Paclitaxel in the branches and leaves,under the action of chloroform:ethyl acetate : methanol(88: 7: 5) expansion agent the shift value of 0.62 has a blue black point in parallel with the standard point,thus paclitaxel components present in the sample;The third chapter is about the optimization of experimental of extraction process,through the comparison of the single factor and orthogonal experiments,and considering the heat sensitive of taxol and the industrial production needs,the final determination of the optimal extraction technology is premise with ethanol as the best solvent,the extraction temperature is 45℃,the extraction time is 3 hours,the ratio of material to liquid is 1:20,two stage extraction;The fourth chapter is about the quantitative experiments of taxol in the foliage,the taxol concentration of standard solution is in 10.6~106.0μg/ml,the linearregression equation is Y=15.554X-32.882, the correlation coefficient is R =0.9998(n=6), the results show the good linear relationship, The measured average taxol content of Taxus madia in the branches and leaves not after impurity removal process is 0.0312%,The measured average taxol content of Taxus madia in the branches and leaves after impurity removal process is 0.0114%.
