Taking 5-HT, NE in the hypothalamus of rabbits as measure index, the thesis aims at revealing the central mechanism of cheek acupuncture analgesia effect and providing a theoretical basis for the clinical treatment of pain with cheek acupuncture。Methods: 40 rabbits were randomly divided into A, B, C, D groups with excel2003, each group has 10 rabbits, group A is regarded as the normal group, group B as model group, group C as body acupuncture group and group D as cheek acupuncture. The emulsifier is made by mixture of the egg protein powder with 4mg / ml concentration and an equal dose of Freund’s complete adjuvant(CFA). Group B: 27 days after experiment, has the same fix method with other groups for 30 mins every day. Then added normal saline to make into hypothalamus homogenate with 10%, homogenate were put into cold centrifuge 3 000 r / min centrifuged 15 min, the supernatant is set to join the centrifuge tube and stored at-80 ℃ refrigerator. When using high-performance liquid chromatography with fluorescence detection hypothalamus acupuncture pain threshold determination curve peak point serotonin(5-HT), noradrenaline(NA) content.Results:(1) when the buccal group and body acupuncture group were treated with needles for the first time, the analgesic effect changes with time(F=15.780,P= 0.000), there is significant differences in the trend of changing.(F=16.125,P= 0.000)(2)when the buccal group and body acupuncture group were treated with needles for the 10 th time, the analgesic effect changes with time(F=29.378, P= 0.000), there is significant differences in the trend of changing.(F=19.173, P= 0.000)(3) there is no statistical difference in the trend of analgesic effect of acupuncture in the first and the tenth times treatment for cheek acupuncture group(F =0.973,P=0.486).(4) there is no statistical difference in the trend of analgesic effect of acupuncture in the first and the tenth times for body acupuncture group.(F = 2.750, P= 0.064).(5) After the acupuncture treatment, the 5-HT content in the hypothalamus in the cheek acupuncture group and body acupuncture group were significantly increased compared with the model group, there is statistical significance(P<0.05), the ratio of NEand 5-HT in the cheek acupuncture group and body acupuncture group were decreased compared with the model group, there is statistically significant(P<0.05)Conclusions:(1) Cheek acupuncture Analgesia has obvious time curve, after 5mins treatment there is obvious analgesic effect, there is peak at 30 mins, and minimum at 240 mins.(2) Cheek acupuncture group and body acupuncture group has the similar Time-effect Relation of analgesic effect, but there are significant differences。 The analgesic effect of cheek Acupuncture group has the instant effect, rising and decreasing quickly; while the body acupuncture group has the slow effect, rising and decreasing slowly.(3) Both cheek acupuncture and body acupuncture can affect 5-HT and NE contents in the rabbit hypothalamus, it make 5-HT levels were significantly increased and NE / 5-HT ratio decreased, there is no statistical difference. |
