Objective To observe the treatment of Bone Marrow Stem Cells(BMSCs) transplantation by vein and investigate the differentiation of them transplanted in the hepatic tissue of acute liver injury mice, and study the mechanism of BMSCs transplantion on the treatment of acute liver injury in transcriptional activation and m RNA splicing.Methods The female kunming mice were randomly divided into normal control group, damage model group and stem cell transplantation group. The BMSCs proliferated in vitro were labelled with 5-bromo-2-deoxyuridine(Brd U) and transplanted by tail vein injection into the isogeneic acute liver injury mice which received gavage with 40%CCl4. At the fifth day, the tenth day and the fifteenth day after damaged, the livers were extracted from mice in damage model group and stem cell transplantation group. The liver in normal control group without any treatment extracted the livers directly. The liver continuous freeze sections,paraffin sections and fresh tissues were obtained. The liver morphologic changes were identified by hematoxylin-eosin staining, The distribution of the Brd U positive transplanted cells were detected by immunohistochemistry. The expression of NF-κB and CUGBP1 were detected by immunohistochemistry and western blot.Results(1) HE staining results showed that at the fifth day, the tenth day and the fifteenth day after damaged, the repair status of liver tissues in stem cell transplantation group were much better than that at the same time in the damage model group. At the fifteenth day, the liver tissues of stem cell transplantation group were close to normal.,(2) Immunohistochemical results showed that the Brd U positive cells were with yellow or brown nucleus. At the fifth day after damaged, Many Brd U positive cells could be observed scattered in the liver. At the tenth day after damaged, the number of Brd U positive nucleus were fewer than that at the fifth day. At the fifteenth day after damaged,Brd U positive cells were decreased significantly.(3) Immunohistochemical results showed that NF-κB and CUGBP1 in nomal control group was a little expression or none expression in the cytoplasm. Compare with the nomal control group mice, the positive expression degree of NF-κB and CUGBP1 were all increased. Comare with the damage model group, the expression degree of NF-κB and CUGBP1 in the stem cell transplantation group were all decrease, have the remarkable difference in statistics. Western blot results showed that compare with the nomal control group mice, the expression level of NF-κB and CUGBP1 were all increased. Comare with the damage model group, the expression degree of NF-κB and CUGBP1 in the stem cell transplantation group were all decrease, At the same day, positive cells were decreased significantly. Conclusion BMSCs by vein transplantation could migrate into the isogeneic dameged liver of mice and promote the repair process. and over time the number of stem cells per unit area decreased. The treatment of BMSCs in acute liver injury may be related to the decreased expression of NF-κB and CUGBP1, we speculated that BMSCs could decreased the activation of NF-κB and CUGBP1 to reduce the inflammatory reaction and stable m RNA,thus to promote the repairment of the injury liver.
