The Effect Of MiR-20a On ABCA1 Expression And Athe Rosc LeRotic Lesion In Apolipoprote In E Knockout Mice

Objective:We regard apo E knockout(apo E-KO) mice as the research object,to fed them with high fat diet(HD) induced atherosclerosis, to observe the effect of mi R-20 a on ABCA1 Expression and Atherosclerotic lesion.Methods:1.30 male apo E-/-mice of 6 week old were fed a high- fat diet(21% fat, 0.21% choles terol)for 10 weeks,then were randomly divided into 3 groups. 2.Experimental groups: ① T he PBS group:to inject 10 mg/kg PBS(The same dose as mi R-20 a group)by intraperito neal, having twice injections in the first week, then injected once a week for 3 weeks; ② T he increase of mi R-20 a group: to inject mi R-20 a agomir 10 mg/kg(diluted with DEPC wa ter), injections method is the same as the the PBS group;③The inhibition of mi R-20 a group: To inject mi R-20 a antagomir 10 mg/kg(diluted with DEPC water), injections metho d is the same as the PBS group. The Detection Index:1.After 4 weeks, to measure HDL-C、LDL-C、TG、TC of every group;2.To detect the expression of liver ABCA1 protein using Western blot; 3. Using the Real- Time PCR to detect the expression of liver ABCA1 m RNA and the content of mi R- 20 a;4.To observed the atherosclerotic lesions area of aortic root.Results:1.Compared with the PBS group, after the mi R-20 a agomir intervention, serum HDL-C decreased,LDL-C and TC increased,and TG is almost the same as PBS, mi R-20 a expression increased,ABCA1 m RNA and ABCA1 protein expression of liver decreased, and atherosclerotic lesions aggravated.2. Compared with the PBS group, after the mi R-20 a antagomir intervention, serum HDL-C increased, LDL-C and TC decresed,TG is almost the same as PBS, mi R-20 a ex pression decreased,ABCA1 m RNA and ABCA1 protein expression of liver increased, and atherosclerotic lesions reduced.Conclusions:We identified anti- mi R-20 as a novel mi RN A to decrease the atherosclerotic lesions area,it expected to become a new type of anti atherosclerosis drugs.
