| Objective:(1).By observing Th17/Treg cells and IL-23/IL-17 of AS to find whether AS disease activity and pathogenesis are related to IL-23/IL-17 and Th17/Treg.(2).By observing Th17/Treg cells and IL-23/IL-17 after methylprednisone treatment of AS,find the influence of methylprednisone.Methods:Selected 30 patients with active AS as subjects, assess the indicators of BASDAI,BASFI, the pain at night,ESR,CRP,according to the clinical symptoms and disease activity of patients, were given short-term high-dose MP treatment,2.5~4mg/kg,Qd,one course was3~4d,interval 3~4d,2~3 courses.Using FCM test Th17/Treg,at the same using PCR determination of IL-23 P19 m RNA IL-17 A m RNA expression level of AS in pretherapy and post-treatment,find its relevance on clinical effect.Results:The AS patients Th17 cells,IL-23 and IL-17 expression levels on peripheral blood were higher,but the normal not, the BASDAI and so on were correlated,Treg is lower,but negatively with above indexes.Th17 IL-23 and IL-17 have relationed.Th17 IL-23 and Treg have negatively correlated.After treatment of the AS,the indicators of BASDAI,BASFI,the pain at night,blood sedimentation,CRP are decreased.After MP treatment,AS PBMCsTh17,IL-17 with IL-23 expression are lower than before treatment,but the distributionabout Treg are more than before treatment.Conclusions:The AS’disease activity is related to IL-17/IL-23 and Th17/Treg,IL-23//IL-17 and imbalance of Th17 and Treg cells may be involved in the pathogenesis of AS.MP can down regulation Th17 cell function, increase Treg function, inhibit IL-23 and IL-17,and can significantly improve joint symptoms and disease activity in patients with AS. |
