To investigate the relationship between the expression of Pin1 and Cyclin D1 and clinical significances of gastrointestinal stromal tumors.Methods:1. 85 cases of gastrointestinal stromal tumors tissues paraffin-embedded specimens and 15 cases of adjacent non-cancerous tissues were collected after operation from Fu Jian Medical University Union Hospital in 2007 to 2014. The protein expression of Pin1 and Cyclin D1 were detected by immunohistochemistry.2. 40 cases of gastrointestinal stromal tumors tissues paraffin-embedded specimens and their adjacent non-cancerous tissues were collected after operation from Fu Jian Medical University Union Hospital in 2007 to 2014. The m RNA expression of Pin1 and Cyclin D1 were detected by Real-time Quantitative polymerase chain reaction.3. To analyse the relationship between the expression of Pin1 and Cyclin D1 and clinical pathological parameters of gastrointestinal stromal tumors.4. To evaluate the correlation between the expression of Pin1 and Cyclin D1 in gastrointestinal stromal tumors.Results:1 Immunohistochemistry results1.1 The expression of Pin1 was located in the cell nucleus and cytoplasm in gastrointestinal stromal tumors tissues and adjacent non-cancerous tissues. The positive expression of Pin1 in gastrointestinal stromal tumors tissues and adjacent non-cancerous tissues was 64.7%(55 /85) and 26.7%( 4 /15),which was statistically significant(p<0.05). And there is no obvious correlation between the expression of Pin1 protein and the genderã€ageã€mitotic figureã€the grade of NIHã€tumor diameterã€tumor locationã€histological type and distant metastasis of patients(all P>0.05).1.2 The expression of Cyclin D1 was located in the cell nucleus and cytoplasm in gastrointestinal stromal tumors tissues and adjacent non-cancerous tissues. The positive expression of Cyclin D1 in gastrointestinal stromal tumors tissues and adjacent non-cancerous tissues was 42.3%(36 /85) and 6.7%( 1 /15),which was statistically significant(p<0.05). The expression of Cyclin D1 protein was not related to the genderã€ageã€mitotic figureã€tumor locationã€histological type and distant metastasis of patients(all P>0.05), but correlated with the grade of NIH and tumor diameter(p<0.05).1.3 There was a significant correlation between the expression of Pin1 and Cyclin D1 in gastrointestinal stromal tumors(r=0.284,p<0.01).2 q RT-PCR results2.1 The CT values of Pin1 in 40 cases of gastrointestinal stromal tumors tissues and adjacent non-cancerous tissues were 6.02±1.13 and 8.94±1.56, a statistically significant difference( p < 0.05).The expression of Pin1 m RNA in gastrointestinal stromal tumors tissues was 7.03 times that of adjacent non-cancerous tissues. And there is no obvious correlation between the expression of Pin1 m RNA and the genderã€ageã€mitotic figureã€the grade of NIHã€tumor diameterã€tumor locationã€histological type and distant metastasis of patients(all P>0.05).2.2 The CT values of Cyclin D1 in 40 cases of gastrointestinal stromal tumors tissues and adjacent non-cancerous tissues were 6.02±1.13 and 8.94±1.56, a statistically significant difference(p<0.05).The expression of Cyclin D1 m RNA was not related to the genderã€ageã€mitotic figureã€tumor locationã€histological type and distant metastasis of patients(all P>0.05), but closely correlated with the grade of NIH and tumor diameter(p<0.05).2.3 There was a significant correlation between the m RNA expression of Pin1 and Cyclin D1 in gastrointestinal stromal tumors(r=0.337,p<0.05).Conclusion:1. The expression of Pin1 and Cyclin D1 in gastrointestinal stromal tumors was increased. And they may play a significant role in carcinogenesis and development of gastrointestinal stromal tumors.2. There is no obvious correlation between the expression of Pin1 and the genderã€ageã€mitotic figureã€the grade of NIHã€tumor diameterã€tumor locationã€histological type and distant metastasis of patients.3. The expression of Cyclin D1 was not related to the genderã€ageã€mitotic figureã€tumor location 〠histological type and distant metastasis of patients, but closely correlated with the grade of NIH and tumor diameter.4.
