| Background:With the upward trend of aging population, Atherosclerosis(AS)whose incidence increases, can result in coronary heart disease, stroke and other peripheral organs dysfunction and so on. At present,the doctrine of inflammation which is one of pathogenesis of Atherosclerosis has gradually attracted the attention of people.Adhesion molecules,including monocyte chemotactic protein-1(MCP-1),intercellular adhesion molecule-1(ICAM-1),vascular intercellular adhesion molecules-1 (VCAM-1) and platelet endothelial cell adhesion molecule-1 (PECAM-1), are closely related to the occurrence and development of atherosclerosis. Some studies have found that the sympathetic nervous system plays an important role in the regulation of renin-angiotensin-aldosterone system (RAAS).RAAS activated by sympathetic overactivity, produces a lot of renin and angiotensin Ⅱ (Ang Ⅱ). At the meantime, Ang Ⅱ plays an important role in the formation of atherosclerosis, which can promote the process of atherosclerosis. Renal denervation (RDN) has made some progresses in the treatment of resistant hypertension and improve heart function, but it’s effections on the adhesion molecules and the related mechanisms in the formation of atherosclerosis is unclear.Objective:To evaluate the effects of the RDN on adhesion molecules in atherosclerosis of rabbits.Methods:Twenty-eight male New Zealand White rabbits were randomly divided into control group, RDN+HFD group, Sham+HFD group and HFD group. Blood samples were collected to measure NE, oxidized low density lipoprotein (ox-LDL) levelsAng Ⅱ,MCP-1 and PECAM-1 were detected by immunohistochemistry. ICAM-1 and VCAM-1 were detected by Western bolt and the mRNA expressions of MCP-1, ICAM-1, VCAM-1 and PECAM-1 were detected by RT-PCR.Results:The NE level was lower in RDN group than that in sham and HFD groups after RDN procedure. Triglyceride(TG) was lower in RDN group than that in HFD group. Plasma ox-LDL was significantly decreased in RDN+HFD group compared with HFD group and Sham+HFD group (P<0.05). Meanwhile, the expressions of Ang Ⅱ, MCP-1, ICAM-1, VCAM-1 and PECAM-1 were significantly down-regulated in RDN+HFD group (P<0.05). the ratio of intima-media thickness in aorta was significantly decreased in RDN group compared with HFD group (P<0.05).Conclusions:RDN could decrease sympatholytic activity and level of TG, down-regulate the expression of adhesion molecules, so it may delay atherosclerosis in cholesterol-fed rabbits. |
