| The heart is the central component of the circulatory system, and one of the most important organs. At present, the heart disease has become the common pathema affecting human life and health greatly. If animal experiment was adapted to research the pharmacology, it had not only the high cost and difficult operation, but also the existence of moral pressure. If computer model was used to simulate the electrophysiological activities, it would provide better security to develop drugs economically. Currently it had been main research object by ion channels change to simulate heart nosogenesis. Genetic forms of the Short QT Syndrome (SQTS) arise due to cardiac ion channel mutations. Although the SQTS is an electrical disorder, the heart is both an electrical and mechanical organ and it is feasible, at least in principle, that abbreviated repolarization in the syndrome might influence the mechanical function of the heart. So this thesis discussed that accelerated ventricular repolarization has potential impact on heart mechanical systole.This thesis used the ten Tusscher, Noble, Noble and Panfilov (TNNP) human ventricular cell model to study. First, simulate action potential for single myocardial cell and build the process of action potential propagation through ID myocardial cells, and calculated the ventricular components of ECG, namely pseudo-ECG. Then, compared with O’Hara-Rudy Dynamic (ORD) model, it has also been suggested to be suited to the study of re-entrant arrhythmias in human ventricular tissue. Because the action potential duration for the mid-myocardium cell is not consistant with clinical result, this thesis developed an improved 1D human ventricular myocytes model.This thesis researched the IKr-channel-linked short QT variant 1 and IK1-channel-linked Short QT variant 3. Respectively, the Markov chain IKr model and the Hodgkin-Huxley IKI model formulations were incorporated into TNNP model to gain the action potential, QT intervals and [Ca2+]i et al. Compared simulation results with clinical experiments, It has been shown that abbreviated repolarization may reduce ventricular mechanical function.With Vistual Studio 2010, this thesis got SQT1 and SQT3 simulation data, which were visualized by Matlab. By analyzing pathological properties, it is shown that the simulation results are consistent with clinical data. |
