Objective: Cervical cancer is one of the most common malignant tumors of the female reproductive system, and is the third on the list of incidence of female cancer. The incidence of cervical cancer is at the top of the female genital tract malignant tumors’ list. In recent years, women sufferring from cervical cancer are becoming younger and younger, which is a serious threat to the health of women.Widely existing among different species of endogenous non-encoding short sequence RNA, micro RNA is involved in the physiological processes such as apoptosis, proliferation and differentiation of the cells through controlling its target genes after the transcription. micro RNA is almost involved in the whole biological processes related to regulation and expression of genes.(Caspase) family is a very important protease in the process of apoptosise, and Caspase3 is the most important one of the apoptotic effect. At present, there are few studies on the association between micro RNA Caspase3 polymorphism and cervical cancer in China and abroad. This study is to explore the association between Caspase3 Polymorphism rs1049216 Single Nucleotide Polymorphism(SNP) micro RNA and susceptibility of patients with cervical cancer.Methods: Collecting the Peripheral blood samples from patients(515cases) with cervical cancer and healthy people(415 controls) respectively, and DNA of peripheral blood was extracted to detect Caspase3 3’-untranslated region(UTR) rs1049216 with polymerase chain reaction- primer introduced restriction analysis, PCR-PIRA.The protein expression of Caspase3 in cancer tissues from 515 cases was determined by immunohistochemical method.All data have been statistically analyzed by SPSS19.0, and the application of chi square test has been used to analyze the different groups(age, tumor stage, HPV infection, lymph node metastasis and so on) P<0.05 had statistical significance.Results:1 Control group and the Group’s age, smoking status, no statistically significant difference(P=0.16). In addition, according to marriage, parity conditions are analyzed, no significant(P=0.16). HPV infection in the case group frequencies significantly higher than the control group( P<0.0001).2 C/C genotype as control, C/C, T/C genotype frequency was not significant,(OR=1.09, 95%CI=0.955-1.276, P=0.133), C/C, T/T allele frequencies exist significantly(OR=0.35, 95% CI=0.154-0.581, P<0.0001).Relative to the c-allele, t allele may significantly reduce the risk of developing squamous cell carcinoma of the cervix(OR=0.86, 95%CI=0.473-0.901,P=0.003), statistics and found to be carrying the c allele(T/T), the risk of cervical cancer was carrying c allele(C/C+T/C) individual 0.31 times(OR=0.31, 95%CI=0.126-0.513, P<0.0001).3 The clinical classification of cervical cancer is analyzed stratifiedly, and T/T genotype can significantly reduce the risk of cervical cancer, compared to C/C genotype, but no significant difference between C/C and C/T genotype.4 The groups are not transfers lymphatic metastasis and, compared with C/C genotypes, respectively, compared with the T/C, T/T genotype, genotype frequencies are both no significant differences(P=0.41; P=0.25).5 Caspase3 protein expression in squamous cell carcinoma of cervix,immunohistochemical staining results visible, positive granules in tumor cells and part of the interstitial cells. Caspase3 rs1049216 gene and protein expression(C/C, T/C and T/T) no correlation.Conclusion:1 Caspase3 gene of 3’-UTR micro RNA combined bit points rs1049216 SNP and cervical cancer Squamous cell cancer easy perceptual of associated sex, and C/C gene type compared, carry T/T gene type can obviously reducedcervical cancer Squamous cell cancer of occurred rate, and carry t, bit gene may on cervical cancer Squamous cell cancer of genetic easy perceptual has protection effect, and and tumor staging and the gynecological tumor family history about.2 Caspase3 3’-UTR of micro RNA genes binding site of rs1049216 SNP correlation with lymph node metastasis in squamous cell carcinoma of the cervix.3 There was no difference in Caspase3 protein expression of tumor tissues among three genotypes(CC, CT and TT).
