Objective:Tested by high performance liquid chromatography (HPLC) method, the best compatibility proportion of Rhizoma Atractylodis Macrocephalae and Radices Paeoniae Alba is determinated. By building irritable bowel syndrome rats model, Pharmacodynamics of Rhizoma Atractylodis Macrocephalae and Radices Paeoniae Alba Herbal Pair in the treatment of irritable bowel syndrome in rats are discussed.Methods:Rhizoma Atractylodis Macrocephalae, radix paeoniae alba and different proportion of Rhizoma Atractylodis Macrocephalae and Radices Paeoniae Alba Herbal Pair, which was extracted by water, was determinated by HPLC. Comparison of compositional change in Rhizoma Atractylodis Macrocephalae, radix paeoniae alba and Herbal Pair of Rhizoma Atractylodis Macrocephalae and Radices Paeoniae Alba, the best compatibility proportion was determinated. Rat model with high sensitivity in the gut was built by neonatal maternal separation plus stimulating rectums of adult SD rats with acetic acid.Experiment was divided into six groups, named with the normal control group, model control group, positive control group, Rhizoma Atractylodis Macrocephalae group, radix paeoniae alba group and Herbal Pair group.Ten rats were in each group.Except the normal and model group were given with the same dose of normal saline, the other groups were given corresponding drugs respectively.Different ways to evaluate the sensitivity in the gut with drug intervention included the abdomen retracement reaction (AWR), serum serotonin (5-HT) content, vasoactive intestinal peptide (VIP) in rat plasma and somatostatin (SS) content determination and mast cells (MC) countResults:The best compatibility proportion of Herbal pair was 2:3. Rat model with high sensitivity in the gut,built by stimulating rectums with acetic acid met the characters of IBS. Compared with the normal group, the AWR value of the other groups was rised(P<0.01) obviously. Compared with the normal group, items were rised, including 5-HT, intestinal sensitivity,SS and mast cell count of VIP. After given with pinaverium bromide, Rhizoma Atractylodis Macrocephalae, radix paeoniae alba and 2:3 Herbal Pair, they were all decreased. Action of Herbal Pair group was superior to Rhizoma Atractylodis Macrocephalae group and radix paeoniae alba group(P<0.01).Compared with positive control group, there was no statistically significant difference (P>0.05).Conclusion:Combination with Atractylodes and radix paeoniae alba paeoniflorin atractylodes lactone I and atractylodes lactone â…¢ were easily extracted, and when the best proportion of Rhizoma Atractylodis Macrocephalae and Radices Paeoniae Alba get to 2:3, dissolution rate of paeoniflorin, atractylodes lactone I and atractylodes lactone â…¢ were highest in aqueous extract. Compared with single solution extract, dissolution rates were respectively 104.2%,232.3% and 232.0%. Herbal pair could reduce 5-HT in serum, contentof VIP and SS in plasma and mast cell count in colon tissue, and reduce intestinal sensitivity, so as to cure the disease.
