Objective:To assess the acute pain analgesic effect of different thermal moxibusiton and whether mediated by TRPV1.Methods:Wild type C57 BL/6J mice (WT) and TRPV1 knockout mice (KO) was divded into WT control group (WT+NS), WT model group (WT+FA), WT model plus 45℃ temperature moxibustion group (WT+FA+45℃) and WT model plus 35℃ temperature moxibustion group (WT+FA+35℃), KO control group (KO+NS), KO model group (KO+FA), KO model plus 45℃ temperature moxibustion group (KO+FA+45 ℃) and WT model plus 35℃temperature moxibustion group (KO+FA+35℃), the "zusanlin (ST36) "on the left hind leg of mice was the acupoint of moxibustion. After moxibutstion, inject 10μL 1% formalin at the bottom of left foot in mice, the control group inject 10μL saline on the same part, and then record the mice licking time in 1 hour.Result:1. The analgesic effect of different thermal moxibustion in acute pain:both different thermal moxibustion (45℃and 35℃) can significantly reduced the formalin pain modle mice licking time in early phase (neuralgia) and late phase (inflammatory pain), and the licking time of moxibustion at higher temperature (45℃) was lower.2. TRPV1 affect acute pain analgesic effect of different thermal moxibustion: the 45℃thermal moxibustion can significantly reduced the formalin pain modle mice licking time in early phaseand late phase both in WT and KO, and the licking time of moxibustion of WT was lower. While the 35℃ thermal moxibustion can significantly reduced the formalin pain modle mice licking time in early phaseand late phase both in WT and KO, and there are no significant differences between WT and KO.Conclusion:l.Both 45℃ and 35℃ thermal moxibustion have analgesic effect of formalin pain, and the analgesic effect of moxibustion at higher temperature (45℃) was better.2.TRPV1 involve in the acute pain analgesic effect of moxibustion in 45℃ thermal moxibustion, and has no effect on 35℃ thermal moxibustion.
