Objective:By comparing normal subjects and in patients with NAFLD leukocyte telomere length, and the relationship between NAFLD liver cell senescence gene P21 between the liver tissue pathology, explore liver cell senescence in NAFLD progression effect. Methods:Study of the relationship between liver aging and nonalcoholic fatty liver disease (NAFLD) we included from January 2015 to March 2016, Affiliated Hospital of Hangzhou Normal University liver disease center in outpatient and inpatient, and (1) with no history of drinking or drinking alcohol equivalent to less than 140g/weeks (70g/< female week); (2) except for viral hepatitis, drug-induced liver disease, total parenteral nutrition, hepatolenticular degeneration autoimmune liver disease, can lead to disease specific fatty liver; (3) pathological liver biopsy histological changes consistent with fatty liver disease diagnostic criteria or radiographic evidence of fatty liver; a total of 108 patients with NAFLD, including 20 cases of liver biopsy; Age is selected in the same period of physical examination population, sex matched healthy controls 99 cases; Detailed inquiry into the crowd disease history and way of life and health. Fasting blood serum separation determination of blood biochemical indexes. The PCR detection of peripheral blood leukocyte telomere length. HE staining evaluation of inflammation and fibrosis, the immunohistochemical expression of P21.Results:Compared with NAFLD group and normal group, NAFLD group of serum liver enzyme index, triglyceride, total cholesterol was significantly higher than the normal group (P <0.001); No significant difference between two groups in age, telomere length is shorter than the NAFLD group and normal group (P<0.05); positive correlation P21 expression in liver cells and liver fibrosis and the degree of fat (R2=0.815, P<0.001 and R2=0.757, P<0.001); The liver inflammation and ballooning have no correlation (R2=0.263, P=0.262 and R2=0.193, P=0.442) had negative correlation with.NAFLD of telomere length of peripheral blood leukocytes and liver fibrosis and steatosis (R2=-0.640, P=0.006 and R2=-0.570, P=0.009). The small leaf and liver inflammation and ballooning no correlation (R2=0.110, P=0.643 and R2=0.021, P=0.936) were analyzed. The expression of NAFLD in patients with telomere length in peripheral white blood cells and liver cells P21, results showed that there was a negative correlation between them (R2=-0.54, P=0.025).Conclusion:1) NAFLD patients peripheral blood human telomerase length was significantly shorter in the normal population; 2) Liver cell aging signs of p21 can according to the extent of delamination in NAFLD liver fibrosis and fat as a variable, but not to judge the inflammation of the liver.3) Peripheral blood leukocyte telomere length can be reflected by changes of liver failure cases of old, it can judge the liver fibrosis and steatosis may is one of the indicators of monitoring of NAFLD patients and prognosis.
