Influence Of Yangzheng Xiaoji Capsule On Expressions Of CD4~+ CD25~+ Foxp3~+ Treg Cells And Related Cytokines Of MFC Tumor Bearing 615

Objective Observe the anti-tumor effect to murine foregastric carcinoma (MFC) Tumor bearing 615 in the Yangzheng Xiaoji Capsule; preliminarily discuss the express influence of Yangzheng Xiaoji Capsule on regulatory T (CD4+CD25+Foxp3+Treg) cells, interleukin-10 (EL-10), transforming growth factor-β1 (TGF-β1) in mice peripheral blood and Foxp3mRNA, scurfin protein in the tumor tissue; in order to further clarify the mechanism of Gastric Carcinoma with Yangzheng Xiaoji Capsule.Methods Take the male 615 mice as object of study. MFC tumor bearing 615 mice models were established by inoculated subcutaneously way, and were randomly divided into 4 groups: model group, Yangzheng Xiaoji Capsule group,5-Fluorouracil (5FU) group, Yangzheng Xiaoji Capsule+5FU group. Each group had 10 mouse, and normal group also had 10 mouse. Observe the growth of tumor in MFC Tumor bearing 615, and the change in diet and weight. after being treated with medicine, Executing 615 mice, stripping and weighing tumor tissue, thymus, spleen, calculating tumor volum and inhibitory rate. Using Flow CytoMetry (FCM) to detect CD4+CD25+Foxp3+Treg cells proportion in mice peripheral blood, using Enzyme Linked Immunosorbent Assay (ELISA) to detect the expression level of interleukin-10 (IL-10) and transforming growth factor-β1(TGF-β1) in mice peripheral blood, using Immune Fluorescence Quantitative RT-PCR to detect the expression level of Foxp3mRNA of transp-lanted tumor tissue, using Western Blot method to detect the expression level of scurfin protein of transplanted tumor tissue.Results 1. Successfully established MFC tumor bearing 615 mice models.2. Yangzheng Xiaoji Capsule could improve the whole condition of MFC Tumor bearing 615, could significantly reduce the weight of spleen belonging to peripheral immune organ of MFC Tumor bearing 615 and had statistical meaning(p<0.05), as well as Yangzheng Xiaoji Capsule +5FU.3. Yangzheng Xiaoji Capsule,5FU, Yangzheng Xiaoji Capsule+5FU both could restrain the growth of transplanted tumor of MFC Tumor bearing 615 and the inhibitory rate respectively were 25.89%、22.29%、50.92%.4. In the process of resistance to gastric carcinoma, Yangzheng Xiaoji Capsule could significantly reduce the express of CD4+CD25+Foxp3+Treg cells,IL-10, TGF-β1 in mice peripheral blood and Foxp3mRNA, scurfin protein in the tumor tissue and had statistical meaning(p<0.05) associated with chemotherapy drug.Conclusions Yangzheng Xiaoji Capsule could restrain the growth of transplanted tumor of MFC Tumor bearing 615, improve the quality of life, especially in the process of resistance to gastric carcinoma associated with chemotherapy drug, and the main mechanism may related to the reduce of expression of CD4+CD25+Foxp3+Treg cells, IL-10, TGF-β1 in mice peripheral blood and Foxp3mRNA, scurfin protein in the tumor tissue to reverse tumor immune escape to improve the immune function.
