| Objective:To explore the effect of FOXQ1 gene on EMT of Co lorectal Cancer SW480 cells and its mechanism.Methods:Short-hairpin RNA (shRNA) of FOXQ1 and Negative Control shRNA were constructed and transfected into SW480 cells w hich express high level of FOXQ1.After the lentivirus with the best s ilencing effect was screened,it would be the only silencing one in fol lowing experiments.Next,the SW480 cells were set as Interfered grou pã€Negative groupã€Control group.Cellular morphology and the junction among cells were examined under the microscope,transwell chambers assayã€adhesion experiments were used to detect the ability of invasi onã€migrationã€homogeneous and heterogeneous cells intercellular adh esion of each group.Meanwhile,qRT-PCRã€Western blot were used for analyzing E-cadherinã€N-cadherinã€Vimentin and MMP-2 expression.Results:The lentivirus with FOXQ1-sh1 successfully silenced th e FOXQ1 gene of SW480 cells.Compared with Negative group and Control group,Interfered group extensive cells polarity and tight juncti on among cells increased;invasion and migration ability were weaken (P<0.05);homogeneous cells intercellular adhesion ability was increa sed and heterogeneous cells intercellular ability was opposite(P<0.05); meanwhile,the expression of E-cadherin declined significantly (P<0.05), the expression of N-cadherinã€Vimentin and MMP-2 rose significantly (P<0.05).Conclusion:Silencing FOXQ1 gene can reverse the EMT of the colorectal cancer SW480 cells and inhibit cell metastasis,its mechani sm may be related to the up-regulation of E-cadherin and down-regul ation of N-cadherinã€Vimentin and MMP-2. |
