Background non-small cell lung cancer (NSCLC) is the leading cause of cancer-related deaths worldwide. Stereotactic radiotherapy is suitable for patients with a more limited number of metastatic lesions and recurred, and overall treatment effect is still unsatisfactory. Some reported that glutathione-S-transferases n has high expression in many tumors, which has closely related with diagnosis and treatment of tumor. Recently, more attention has also been focused on anthocyanidin, research has shown that anthocyanidin would antioxidation and be a effective natural free radical scavenger. furthermore, anthocyanidin may reduce the tumor resistance. So, it plays an important role in treatment for tumor. Fewer reports reported effect of anthocyanidin in NSCLC.Objective This study aims to searh a new treatment for NSCLC, to investigate the effect anthocyanidin to the expression of GST-π in NSCLC and to expore its possible mechanism. Moreover, Screen the optimum anthocyanidin concentration for the treatment and clinical research of NSCLC.Methods1 Sampling,separation and cell culture:40 clinical NSCLC specimens, including 20 lung squamous cell carcinomas and 20 lung adenocarcinomas, these clinical NSCLC specimens were isolated and cultured using tissue explant method,and develop to P2.2 Chemosensitivity test:All NSCLC cell were divided into 0μg.1-1group,10μg/ml group, 100μg/ml group,500μg/ml group and 1000μg/ml group according to the concentration of anthocyanin. 0μg/ml group was be control group. The optimal concentration of anthocyanins of NSCLC cell culture medium were investigated by MTT method.3 100μg/ml is the optimal concentration of anthocyanins of NSCLC cell culture medium,so 100μg/ml group was be experimental group,and 0μg/ml group was be control group.The expressions of GST-π protein in two groups were detected by SP immunohistochemistry,the content of GST-π protein in NSCLC cells of two groups were detected by cell-ELISA,and the level of GST-π mRNA in NSCLC cells of two groups were detected by reverse transcription-polymerase chain reaction.Result①100μg/ml anthocyanins for NSCLC cell inhibitory rate is 65%, the rate is higher than the other three anthocyanin concentration, this phenomenon shows that 100μg/ml is the optimal concentration of anthocyanins of NSCLC cell culture medium.②The expressions of GST-π protein in experimental group is lower than that of control group. There were no difference between the expressions of GST-π protein in lung squamous cell carcinomas and lung adenocarcinomas of experimental group. There were no difference between the expressions of GST-π protein in lung squamous cell carcinomas and lung adenocarcinomas of control group.③The content of GST-π protein in lung squamous cell carcinomas and lung adenocarcinomas of experimental group was lower than that in lung squamous cell carcinomas of control group. There were no difference between the content of GST-π protein of lung squamous cell carcinomaswith that of lung adenocarcinomas in experimental group, there were no difference between the content of GST-π protein of lung squamous cell carcinomaswith that of lung adenocarcinomas in control group.④The level of GST-π mRNA in NSCLC cells of experimental group is lower than that of control group, there was no difference between the level of GST-π mRNA of lung squamous cell carcinomaswith that of lung adenocarcinomas in experimental group, there were no difference between the level of GST-π mRNA of lung squamous cell carcinomaswith that of lung adenocarcinomas in control group.Conclusion 100μg/ml is the optimal concentration of anthocyanins of NSCLC cell culture medium, anthocyanin can reduce the expressions of GST-π in NSCLC cell,this can Help to clinical choose effective chemotherapy.help to effective chemotherapy in clinical predictions.
