| Objectives.To perform a comparison of two methods of partial bladder outlet obstruction in the male rat----retropubic and perineal approach expore a high survival rate,reproducibility and stability animal model.It provides a tool to deeply study the mechanism of bladder remodeling.Methods.Eighty healthy male Wistar rats were studied and randomly divided into three groups. Twenty(Group â… ) were selected for a sham operate through a perineal incision.Thirty(Group â…¡) were selected for midprostatic obstruction using a retropubic approach,and thirty(Groupâ…¢) were seclected for bulbous urethral obstruction through a perineal incision. According to duration of obstruction,three groups were respectively and randomly grouped Group-2-week and Group-4-week.After 2-week and 4-week respectively,all experimental animals were anesthetized and underwent cystometric evaluation.Then the animals were killed,and the organs including bladder and seminal vesicle from each of the groups.Bladder body were weighted and recorded,and then partial bladder body and seminal vesicle were fixed in 10%formalin.Remaining bladder body were saved the liquid nitrogen.Results.The survival rate of Group-â…¡ and Group-â…¢ was 73.3% and 80.0%,there is on statistical difference.Operating time in three groups were (9.75(2.29)〠(17.33(3.54)ã€(10.77(2.44) min,respectively.Operating time of Group-â… and Group-â…¢ were lower than Group-â…¡.DLPP of Group-2-week and Group-4-week of Group-â… ,Group-â…¡ and Group-â…¢ were (26.31(2.32)ã€(27.34(3.93)ã€(24.68(2.39) mmHg and (26..42(2.41)ã€(34.23(3.01)ã€(32.63(3.20)mmHg.On 4 weeks,Group-â…¡ and Group-â…¢ were higher than sham group.Group-4-week of Group-â…¡ and Group-â…¢ were apparently higher than Group-2-week.Bladder wetweight of Group-2-week and Group-4-week of Group-â… ,Group-â…¡ and Group-â…¢ were (0.1486(0.0200)〠(0.1881 (0.0490)ã€(0.1797(0.0286)g and(0.1478(0.0236). (0.2567(0.0476)ã€(0.2453(0.0378) g,On 4 weeks,Group-â…¡ and Group-â…¢ were higher than sham group.On 2 and 4 weeks,there is a significant difference. Conclusions.The retropubic and perineal approach can successfully establish a partial bladder outlet obstruction in the male rat. Compared wiht the retropubic approach,the perineal approach have a high survival rate,eproducibility and stability animal model,and that better mimic progress of human male partial bladder outlet obstruction. |
