Objective:To detect the expression of human telomerase reverse transcriptase(hTERT) mRNA in the melanoma,analyzing the relationship and significance between the expression and different subtypes,clinical pathological features of melanoma. Methods: Collected 64 paraffin-embedded tissues of melanomas and 30 normal nevus tissues diagnosed in Xinjiang autonomous region people’s hospital from January 2010 to December 2014. Expression of hTERT mRNA was detected with real-time quantitative PCR in 64 cases of melanoma,30 cases of nevus;Using SPSS17.0 software, to analysis the relationship between hTERT mRNA expression in melanoma and clinical pathological features such as age, gender,ethnicity, different subtypes, lymph node metastasis, Clark classification, etc. Results: In 64 melanomas,27 cases occurs in acral, 23 cases in mucosal and 14 cases occurs in non acral.30 cases are Han,34 cases are Uygur.Using 2-△△ct methods to calculate that the expression of hTERT mRNA in tissue of melanoma groups was 52.43 ± 5.42, significantly higher than that in nevus groups(21.38 ± 3.73)(P<0.05);The expression of hTERT mRNA in melanoma patients was no significant correlation with clinical pathology characteristics such as age,gender,ethnicity(P>0.05),but with different subtypes,metastasis of lymph node,Clark classification(P<0.05).h TERT mRNA expression level in mucosal melanoma was significantly higher than that of acral and non acral melanoma, while the expression level of acral and non acral melanoma was no differences(P < 0.05). Conclusion: The quantitative levels of hTERT m RNA are significantly higher,especially in mucosal melanoma groups;the expression of hTERT m RNA in melanoma was associated with different subtypes, the depth of invasion and lymph node metastasis, suggesting that hTERT perhaps plays an important role in the occurrence and development of melanoma,and to provide clues for the diagnosis, treatment and prognosis of melanoma.
