| Phosphatidylinositol is a glycerophosphatide compound with inositol. PI are mainly used in the pharmaceutical industry to research affecting their metabolism in the cell signal transduction.In this paper, the main purpose is to obtain pure phosphatidylinositol from soybean phospholipids through separation and purification, and to investigate its effect on hypolipidemic rats.In this study, phosphatidylinositol was detected by high-performance liquid chromatography, Si column, mobile phase n-hexane, isopropanol and acetic acid, detection wavelength 208nm. Result shows the purification of phosphatidylinositol in soybean phospholipids was 29%.Use single factor experiment and response surface experiments to optimize the separation conditions. The optimum separation conditions:ethanol concentration 56%,, n-hexane:ethanol (1:1.7, V/V), pH8.17, extracted three times. Purify the crude phosphatidylinositol by metal ion precipitation (Adding 0.3g/LCa2+). Finally, purity of phosphatidylinositol is 84.17%.Male rats were randomly divided into six groups, named blank group, model group, low phosphatidylinositol group(105mg/kgBW·d), middle phosphatidylinositol group (210mg/kgBW·d), and high-dose phosphatidylinositol group(420mg/kgBW·d). The positive control group fed phosphatidylcholine 210mg/kgBW per day. Raising eight weeks, and related indicators were measured.Experimental results show that phosphatidylinositol can significantly reduce the concentration of hypolipidemic rats serum triglyceride (p<0.01), total cholesterol (p<0.01) and non-high-density lipoprotein cholesterol (p<0.01) compare to the model group. In the mean time, the lipid levels composite indicator TG/HDL-C and atherogenic index AI were significantly reduced in high-dose group.In high-dose group, TG and TC concentration in rat liver were significantly reduced (p<0.01), which shows that phosphatidylinositol had a significant effect on lipid metabolism in rat liver. In serum and liver, increasing the doses of phosphatidylinositol, SOD activity increased significantly (p<0.05, p<0.01), choline ester enzyme activity increased significantly (p<0.01), too, but MDA concentration reduced significantly (p<0.05, p<0.01). These results proved the phosphatidylinositol can alleviate lipid peroxide production and protect the liver.By HE staining the liver tissue sections of rats, we found phosphatidylinositol can effectively improve the corresponding pathological symptoms.Using PC as control group, the result is that PC can more effecitely improve the physiological indicators of rats than PI.Through all experiments, phosphatidylinositol can reduce blood lipids, but the effect is not better than PC. |
