| Objective: To investigate the immune regulatory function of Qiamuguer(QMG) on immune suppression mice, analysis the serum characteristic metabolites in immune suppression mice by using 1H NMR-based metabonomics and explore the metabolic mechanism of QMG’s immune regulatory effects. Methods: sixty-five male Kunming(KM)mice were randomly divided into five groups(normal group, Cy-modeled group, Chinese medicine group, QMG high-dose group and low-dose group). Four of them except the normal group were administered Cy(50 mg/kg body weight once a day by intraperitoneal injection) to induce immune suppression. The QMG high-dose and low-dose groups were received QMG treatment(0.374 g,0.1g orally once a day). The Chinese medicine group was orally infused with Zhenqifuzheng granule( 0.133 g once a day). After observing for30 days, we measured the routine blood tests(WBC, Lym, RBC, PLT),T lymphocyte subgroups(percentage of the CD4+T cells and CD8+ T cells, CD4/CD8) in peripheral blood and proliferation of lymphocyte T in spleen, thymus gland index, spleen index,value of IL-2,IFN- γ in serum,value of IgG,IgM,IgA in serum, NMR technology had been used to find out the variability serum metabolites by the method of OPLS-DA.Result: Comparing with the normal group, Cy-modeled group showed a significant decrease in the number of WBC, Lym, RBC, PLT, HGB, percentage of the CD4+T cells, ratios of CD4/CD8 and proliferation of lymphocyte T in spleen, thymus gland index, spleen index, value of IL-2,IFN- γ in serum and value of IgG, IgM, IgA in serum. But both of QMG dose groups and Chinese medicine group showed similar results with normal group on above quota except the PLT, IgM and thymus gland index, which had great difference with the Cy-modeledgroup. 1H-NMR tests showed that Comparing with the normal group, Cy-modeled group showed a significant decrease in the serum level of glutamine, glycolprotein, unsaturated lipid, VLDL, LDL, acetone and showed a significant increase in the serum level of leucine,alanine, tyrosine, histidine, lactate, glycine, creatine, metheionine, valine, β-hydrocxy butyrate and carnitine; QMG high- dose group showed a significant increase in the serum level of alanine, and showed a significant decrease in the serum level of methionine,β-hydrocxy butyrate and Citric acid,which had great difference with the normal group;Comparing with the Cy-modeled group,QMG high- dose group showed a significant decrease in the serum level of inositol, tyrosine, threonine, methionine, histidine, creatine,valine, β-hydrocxy butyrate, Lactic acid, and showed a significant increase in the serum level of acetone, VLDL, LDL, unsaturated lipid, which had great difference with the Cy-modeled group.Conclusion:QMG markedly improve the immune function in immune suppression mice induced by Cy. Immune suppression mice have a imbalance in metabolic network, these findings indicated that lipid metabolism-related metabolites are decreased,however, β-oxidation of fatty acid, proteolysis and glycolysis level are increased. Immune suppression mice after the accepting of QMG intervention, β-oxidation of fatty acid,glycolysis and lipodieretic level are decreased, protein synthesis level are increased,consequently improve the energy metabolism, immune function, integrity of the tissue cells, normal physiological function and structure of tissues in immune suppression mice induced by Cy. |
