| Psoriasis is an immune-mediated skin disease with chronic inflammatory, characterized by epidermal hyperpfoliferation, aberrant differentiation and persistent inflammatory. Studies shows that psoriasis is related to respects, with genetic factors,immunology,neuropsychiatric factors and environmental factors included, while the precise pathogenisis was in the mist. Traditional Chinese medicine has been employed in treatment of a variety of conditions, including skin diseases, with a long history. Zanthoxylum bungeanum(Rutaceae), an important Chinese herb medicine, shows a wide range of biological activities in anti-tumor, anti-inflammatory, and anti-microbial obvious effectiveness on some ailments, such as vomiting,toothache and stomachache, while no published essays reported about anti-psoriasis activity of Zanthoxylum bungeanum essential oil.This study aimed at investigating the mechanisms of anti-proliferative and apoptosis induction of Zanthoxylum bungeanum essential oil(ZBEO)on HaCaT cells, a model cell employed as carrier cell in present study.1. In SRB test, ZBEO was found to significantly inhibit HaCaT cells proliferation in a time and dose-dependent manner,with an IC50 0.024% for 48 h. 2.2. ZBEO induces S phase and apoptosis in HaCaT cells as Flow cytometry test.3. After ZBEO treatment,HaCaT cells showed typical apoptotic morphologic features in DAPI staining assay.4. Western blot assay revealed that after ZBEO treatment, cleaved caspase-8/9/3, PARP and Bax were up-regulated, while Bcl-2 was down-regulated. Furthermore, in TNF-α induced HaCaT cells,NF-кB pathway tends to be a trend of attenuation with ZBEO treatment,via p-IKKα/β down-regulation and IκB-α up-regulation.5. In vitro anti-inflammatory assay indicated that compared with ASA,ZBEO was more effective in albuminous degeneration inhibition.In conclusion,as revealed in graphic abstract,ZBEO can significantly inhibit HaCaT cells proliferation and induce apoptosis via both intrinsic and extrinsic pathways, and the extrinsic pathway was in major phase with caspases involved. Besides,ZBEO can depress the NF-κB signaling pathway,down-regulating COX-2 expression. In the present study,we conducted research on preliminary study of the mechanism of Zanthoxylum bungealum essential oil treatment for psoriasis,providing theoretical basis in psoriasis pathogenesis research and new therapy development. |
