ObjectiveTo observe the effect of lowering serum uric acid and triglyceride in Tongfeng Ning Capsules, this study, we used 3 different animal models of hyperuricemia Detection the xanthine oxidase activity in animal liver, investigate further uric acid lowering mechanism of Tongfeng Ning capsule. The analgesic and anti-inflammatory effects of Tongfeng Ning Capsules were observed by the method of torsion body and ear swelling in mice. Acute toxicity test was conducted in mice to observe the toxic effect of the capsule, Preliminary evaluation of its’ safety. Through the experimental study of the above several parts, to provide experimental basis for clinical application of Tongfeng Ning Capsules.Methods1.Experimental study on the effect of lowering uric acid, lowering blood fat and the mechanism of lowering uric acid of Tongfeng Ning capsules.1.1 60 Kunming male mice were randomly divided into 6 groups with 10 in each group, administration by gavage for one week,The normal control group was injected with normal saline for 2 hours after the end of the last administration, and the other groups were given intraperitoneal injection of Oteracil potassium(300mg/kg).Get blood by picking eyeballs one hour after administration, Detection of uric acid and triglyceride levels in serum, mice were dissected and get the liver tissue, The tissue were homogenated, centrifuged and the supernatant was got, test the level of uric acid and the activity of xanthine oxidase in liver. To observe the effect of Tongfeng Ning capsule in lowering uric acid and lowering blood fat on Hyperuricemia mice and a preliminary study on the mechanism of lowering uric acid.1.2 60 Difake male Quails were randomly divided into 6 groups with 10 in each group, The normal group common quail fed food, The rest group gavage yeast dry powder in the morning, Stomach filled with Tongfeng Ning Capsule in the afternoon,all Quails freely take a meal and drinking. After 4 weeks of feeding,The drugs were administrated by ig for 4 weeks. Experimental study on analgesic and anti- inflammatory effects of Tongfeng Ning capsule2.1 50 Kunming male mice were randomly divided into 5 groups with 10 in each group,intragastricall administered for seven days, 0.5 hours after the last delivery,Intraperitoneal injection of 0.9% acetic acid in each group(0.1ml/10g), Mice were observed for 15 minutes. Mice were observed for 15 minutes to get it twisting times, compute the percentage of analgesia, To observe the analgesic effect of Tongfeng Ning capsule.2.2 50 Kunming male mice were randomly divided into 5 groups with 10 in each group,intragastricall administered for seven days, 0.5 hours after the last delivery,Xylene(2ml) evenly on each mouse ear inflammation induced by both sides, Left ear was not applied as control group. Mice were sacrificed after 1h, Two ears were cut off along with Mice ear baseline, Use diameter 8mm hole punch lay round ear piece in the same part of the two ears, Weight calculation of swelling and inhibition rate, To observe the anti-inflammatory effect of the capsule of gout To observe the anti-inflammatory effect of Tongfeng Ning capsule.Results1.Experimental study on the effect of reducing uric acid and lowering blood fat of Tongfeng Ning Capsule1.1Effect of TFN capsule on hyperuricemia model in mice: Compared with model group TFN low, medium and high dose groups of serum and liver tissue uric acid levels were significantly decreased,The difference was statistically significant(P<0.05), Compared with the model group, serum triglyceride levels in the middle and high dose group of TFN were significantly decreased, The difference was statistically significant(P<0.05 or P<0.01). Low dose group compared with model group, there was a trend of decrease, The difference was not statistically significant(P>0.05). Cmpared with the model group Liver xanthine oxidase activity showed significantly decreased, The difference was statistically significant( P<0.05 or P<0.01)1.2 Effect of gout capsule on hyperuricemia model of quail:The first week of filling the stomach,The serum uric acid level in each dose group of TFN showed a downward trend,The difference is not significant(P>0.05). Fron the second week, There was a significant decrease in serum uric acid levels in each dose group. The difference was statistically significant(P<0.05). Compared with the model group,TFN low and high dose group the serum triglyceride level was significantly decreased after the second week,The middle dose group in first week serum triglyceride levels had decreased significantly, The difference was statistically significant(P<0.05). Compared with the model group the liver uric acid and xanthine oxidase activity each TFN group showed a significant decrease, The difference was statistically significant(P<0.05 or P<0.01).1.3 Effect of Tongfeng Ning Capsule on rat model of hyperuricemia: The rat during sugar feeding,, The level of serum uric acid and triglyceride were compared with normal group were significantly increased, The difference was statistically significant(P<0.05 or P<0.01). Intragastric administration of TFN in the first week, each dose group of serum uric acid levels decline,The difference was not significant(P>0.05). Form the second week, The level of serum uric acid in each TFN group compared with model group was significantly decreased. The difference was statistically significant(P<0.05 or P<0.01),Second weeks of administration of TFN in the middle and high dose group of serum triglycerides compared with model group were significantly decreased, The difference was statistically significant(P<0.01). The serum triglyceride level in low dose group after 4 weeks of treatment were significantly decreased, The difference was statistically significant(P<0.01). Liver tissue levels of uric acid and xanthine oxidase activity compared with the model group showed significantly decreased, The difference was statistically significant(P<0.05 or P<0.01).2.Tongfeng Ning capsule analgesic and anti-inflammatory effect of experimental study2.1Tongfeng Ning capsule analgesic experiment: Compared with the model group rats TFN capsule middle、high dose group writhing significantly decreased, The difference was statistically significant(P<0.05). Compared with model group the low dose group has no significant decrease, The difference was not statistically significant(P>0.05).2.2 Tongfeng Ning capsule anti-inflammatory experiment: TFN capsule middle and high dose group compared with the model group, the mice ear swelling was significantly decreased, The difference was statistically significant(P<0.05). Compared with model group the low dose group has no significant decrease, The difference was not statistically significant(P>0.05).3. Experimental study on acute toxicity of Tongfeng Ning capsule: Acute toxicity test of mice, no one died, The maximum dose reached 444 times of the clinical dose for adults, Observed after administration for 14 days, In the process of experiment, no mice died.Conclusion:1. For three different animal models of hyperuricemia. Tongfeng Ning Capsule has the effect of lowering serum and liver uric acid level. For mice, the effect of lowering uric acid was best at high doses. And the effect of the best on the quail and the rat is the middle dosage. Tongfeng Ning Capsule also has the effect of reducing serum triglyceride level. Experimental study shows that TFN capsule can reduce serum uric acid with it can reduce the activity of xanthine oxidase in experimental animal have a close relationship.2. TFN Capsule the middle and high dose has certain analgesic and anti-inflammatory effect.. Analgesic effect the high doses is better, but anti-inflammatory effect the middle dose is better.3.No acute toxicity was found in the capsule. And the experimental study on the safety of the drug administration is larger,It show that the capsule has high safety factor of clinical application.
