In recent years, people are constantly increase research and development efforts in the aspects of anti-inflammatory hemostasis herbal, it is particularly important to select an appropriate and effective herbal additives and to evaluate the hemostatic antibacterial activity. There are lots of in vitro evaluation index, Previous study only choose several indexes to test, the results can not fully reflect the hemostatic effect. Therefore, choose a variety of evaluation, screened of a hemostatic activity herbal additives is significantly important if we established the evaluation model of the comprehensive evaluation of each index.6 kinds of herbals, which have an in vitro antibacterial and coagulation activity according to the literature or pharmacopoeia documented reported were tested for their the in vitro coagulation activity and antibacterial activity against oral bacteria, Determination thrombin time (TT), Platelet aggregation rate (PAgR) and adhesion rate (PAdR), activity of Anti-thrombin Ⅲ (AT-Ⅲ), plasma Recalcification time (RT), Minimum inhibitory concentration (MIC), Minimum bactericidal concentration (MBC). Total flavonoids, total saponins, organic acid of various herbs were purified by the macro porous adsorption resin, to explore the possible hemostatic components. Analytic hierarchy process to established an in vitro hemostasis model, add 13 kinds of herbal extracts for the same test, in order to enriched and improved the model of the data base. The model was used to compare the in vitro hemostatic effect of samples.4 kinds of samples were selected to tested the bleeding and coagulation experiments to verified the feasibility of the model.Hemostatic experiment showed that the crude extract compared with saline, TT test found that Panax Notoginseng, Yunnan Baiyao and tranexamic acid with the concentration of 0.1~10 mg·mL-1 has a best promoting effect of thrombin activity; PAgT and PAdT found that Yunnan Baiyao, Platycladus orientalis leaf and tranexamic acid with the concentration of 0.1~10 mg·mL-1 can significantly promoted the platelet aggregation and adhesion; RT test found that Platycladus orientalis leaf, Panax Notoginseng and Yunnan Baiyao with the concentration of 0.1~10 mg·mL-1 significantly shortened RT; AT-Ⅲ test found that Mugwort, Platycladus orientalis leaf and tranexamic acid significantly inhibited the inhibitory effect of AT-Ⅲ on thrombin anti-thrombin. For purification, The improvement of total flavonoids of Mugwort, total flavonoids and organic acid of Honeysuckle can be further shortened TT and RT; The increased total flavonoids of Platycladus orientalis leaf will further promoted the PAdR and PAgR, shortened the TT; Total flavonoids and total saponins increased of Panax Notoginseng only can shortened TT, other indicators showed that no obviously improved on coagulation effect before and after purification test; Pollen Typhae by the extended RT became shortened RT, and other tested showed that after purification the result became no change or worse; All indexes of Agrimony were become worse after purified. The antibacterial test showed that for the crude extract Agrimony had the best antibacterial effect on Streptococcus mutans, MIC and MBC were 125 mg·mL-1; Honeysuckle have the best antibacterial effect on Streptococcus mutans and Porphyromonas gingivalis, MIC were 62.5 mg·mL-1 and 15.625 mg·mL-1, but there is no bactericidal effect; there is no bactericidal effect of Panax Notoginseng also. For purified, Platycladus orientalis leaf on Streptococcus mutans MIC were become 37.5 mg·mL-1; Honeysuckle appeared bactericidal effect on Streptococcus mutans with MBC were 7.813 mg·mL-1; There is no significantly improved antibacterial activities for others. MIC of Honeysuckle extract after purification on Porphyromonas gingivalis were become 3.906 mg·mL-1, and MBC were 37.5 mg·mL-1; MIC of Platycladus orientalis leaf were become 300 mg·mL-1; MIC of Pollen Typhae were become 250 mg·mL-1 and MIC of Agrimony were become 62.5 mg·mL-1; Panax Notoginseng appeared inhibition effect on Porphyromonas gingivalis and the MIC were 250 mg·mL-1. Thirteen kinds of herbs were tested too, With the experimental data of twenty kinds of samples, the model of bleeding was established, Better hemostatic effect:Platycladus orientalis leaf, Yunnan Baiyao, Membranaceus, Miltiorrhiza, Ginkgo biloba leaf, Beautyberry grass, Radix paeoniae rubra, Angelica. Animal experiments showed that 3 kinds of herbs could inhibit the mice bleeding time and clotting time, and achieve hemostasis role compared with the control group. In addition, low dose(5 g crude drug·Kg-1) of Panax Notoginseng and high dose(15 g crude drug·Kg-1) of Radix Zanthoxyli promoted the hemostasis and high dose(10,15 g crude drug·Kg-1) promoted the blood circulation. This is consistent with the model evaluation results.Total flavonoids of Platycladus orientalis leaf and Mugwort, total flavonoids and organic acid of Honeysuckle may the effective components of the hemostatic components; Total flavonoids of Pollen Typhae mainly rely on influenced the endogenous coagulation system to play the role of promoting coagulation; total flavonoids of Agrimony and total saponins of Panax Notoginseng has a potential ability of anti-thrombotic. Total flavonoids of Platycladus orientalis leaf may be the main antibacterial components on Streptococcus mutans; Total saponins of Panax Notoginseng is likely the matter to inhibited Porphyromonas gingivalis; Honeysuckle has a good antibacterial effect on Streptococcus mutans and Porphyromonas gingivalis with its total flavonoids and organic acid; Total flavonoids and other components of Mugwort, Agrimony and Pollen Typhae made a synergistic antibacterial; Mice bleeding and blood coagulation test indicates that the model is basically suitable.
