| Objective: To analyze the chemical composition of latex of Ficus carica from Atush, Xinjiang. Method: 1. Separate and analyze chemical constituents by column chromatography, TLC, HPLC, GC-MS, LC-MS thchniques and identify the structures on the basis of 1D, 2D NMR and mass spectroscopies. Determine the anti-vitiligo activity of main components. 2. Determine the content of three coumarins in Fig latex by HPLC method. 3. Establish HPLC fingerprint and determine coumarins in latex of Ficus carica collected from different locations of Atush. Result: 1. Nine compounds were isolated using various modern chemical separation and analysis techniques,among them, the structures of seven compounds were identified by 1H-NMR, 13C-NMR, 1H-1H COSY, HSQC, HMBC spectra analysis as psoralen, bergapten, umbelliferone, Dibutyl phthalate, Di-2-Ethylhexyl phthalate-D4, 3,4-Epoxide psoralen and 3,4-Epoxide-5-methoxy psoralen. Wherein, the psoralen, bergapten, umbelliferone and ethyl acetate fraction showed anti-vitiligo activity. 2. The method for determine the three compounds of latex of Ficus carica was established. There was a good linear relationship over the concentration range of(0.001~0.019) mg/ml, R2= 0.9999 for umbelliferone,( 0.01~0.19) mg/ml, R2 = 0.9997 for bergapten and(0.01~0.19) mg/ml, R2 = 0.9999 for psoralen. The average recoveries of umbelliferone, bergapten and psoralen in methanol part were 98.09%(RSD = 0.85%) 102.07%(RSD = 1.07%)and 101.00%(RSD = 0.71%); and in ethyl acetate fraction the average recoveries were 100.03%(RSD = 1.72%), 98.72%(RSD = 1.53%)and 98.61%(RSD = 0.67%), respectively. The content of umbelliferone, bergapten and psoralen in methanol part were 1.09, 9.89, 24.58 mg/g, in ethyl acetate fraction were 13.38, 146.70, 282.90 mg/g, respectively. 3. HPLC fingerprint chromatogram of ten batches of Ficus carica L. latex samples were established. All samples contain 13 peaks, and the relative retention time of common peaks is almost same, but the difference of their relative peak area is obvious. Conclustion: 1. This research is conducive to further development and utilization of latex of Ficus carica and to find medical active ingredient. 2.The determination method is simple, accurate and stable. It could be applied to the determination of quality control. 3. This method of fingerprint analysis is stable and should be used for identification and quality assessment of latex of Ficus carica L.. |
