Renal cell carcinoma (renal cell carcinoma, RCC), referred to as kidney cancer, is one of the highest mortality rate of neoplastic diseases of the urinary system, accounting for about adult systemic cancer 2 to 3%, and every year in the 3 to 4% the rate rise, the incidence of cancer in men ranked seventh, ninth in female cancer, and accompanied by younger and younger. An annual increase of kidney cancer worldwide is about 200,000 cases,of which died of kidney cancer are about more than 100,000 cases.When patients diagnosed when about 30% of tumor metastasis has occurred, and transfer of patientswith 2-yearsurvival rate is less than 20%, So the kidney cancer has become an important disease threat to people’s health.Existing studies suggest that kidney cancer originated in the urinary tubule, associated with multiple genes, such as:c-MET, VHL, FHIT, BHD, TSC1, TSC2, EGFR, NRC.l and RASSFIA etc. Depending on the type of renal cell carcinoma, renal cell carcinoma can be divided into clear cell renal cell carcinoma, papillary RCC, chromophobe cell carcinoma, collecting duct carcinoma and undetermined type, clear cell carcinoma is the most common type, accounting for about75%. However, the cause of kidney cancer is unclear.Studies have shown that, von-Hippel-Lindau (VHL) tumor suppressor genes is closely related with occurrence of clear cell renal cell carcinoma.VHL is an important member of the signaling pathways involved in hypoxia-inducible. In the hypoxic tumor microenvironment or mutated VHL, VHL is phosphorylated cause hypoxia inducible factor (Hypoxia inducible factors, HIFs) inhibition of inactivation, HIF promote expression of a downstream, such as VEGF. and enhanced angiogenesis, thereby promoting kidney cancer development. Despite progress with the progress of research on mechanism of renal cell carcinoma, we developed some appropriate therapeutic drugs, such as:anti-angiogenesis drugs target Sorsfenib, Sutent, but in the clinical application and the cure rate is low and generate resistance.. Minimally invasive surgical technique is still the main method of treating kidney cancer clinic, but its presence can significantly recurrence, so now its efficacyremains controversial. Although immunotherapy can enhance the body’s anti-tumor, reduce tumor recurrence, improving patient survival, but because of immune recognition, harming normal tissues and so on, so that the efficacy of cancer immunotherapy is far from satisfactory. Therefore, we should find new drugs to treat renal cell carcinoma, overcome this problem. Research related signaling pathways and components, making new therapeutic targets.Scutellariabaicalensis (Scutellaria baicalensis Georgi), alias camellia roots, soil gold tea root, is a perennial herb, is a common medicine. More and more studies show skullcap has a variety of anti-tumor effect, the active ingredients are gradually separated out and purified. Through screening active compounds in Scutellariabaicalensis by SRB method, we find two new renal cancer cell lines significantly inhibited by Compound 1 (bw-lzj-6504), compound 2 (bw-lzj-6517a), and find their IC50.By using immunoblotting to analysis The expression of protein levels of PARP1、CL-PARP、Caspase3、CL-Caspase3、LC3I、LC31I and P62.It was found that the Compound radix scutellariae brass can significantly inhibit the proliferation of 786-0 and induce apoptosis and autophagy. Thus skullcapflavone compound may inhibit the proliferation of kidney tumor cells by promoting apoptosis and autophagy.
