The Effect Of Qianggubao Decoction On Primary Osteoarthritis Of Knee Model In The Ovaries Rats

ObjectivesEstablishing rats model of primary knee osteoarthritis after ovariectomy and observing effect of Qianggubao on articular cartilage and synovial inflammation and estrogen levels in the model,to provide ideas and evidence for Qianggubao therapy in older women primary knee osteoarthritis.Methods1.The establishing and identification of the rats model:24 SPF grade female SD rats,6 month aged,were randomly divided into normal group(NOR),Runing Group(RUN), Ovariectomized Group (OVX),Model Group,(MOG),6 in each group.The rats in NOR were raised in routine way,in OVX Group were ovarientomized and raised in routine way,in MOG Group on the slop-15°fter ovarientomized.Experimental operation plan:to ovarian scheme adopts line into the womb road,probe the bilateral ovarian tissue along the abdominal wall, ligation of the uterus in ovarian tissue 0.5 cm distance place,cut to the ovary and its surrounding tissues;Treadmill training scheme used to 15°downhill,after the 1 W adaptability training,fixed frequency for 28 m/min,60 min/d,6 d/W,6 W continuous training.After collecting 24 h urine,centrifugal supernatant,abdominal aortic blood,serum,centrifugal after ELISA method to detect the urine and blood in the urine of E2,CTX-II content;Take double knee joint femoral, condyles line of pathological tissue section,by HE staining,light microscope under the cartilage Mankin marking,observe morphological changes of articular cartilage,appraisal model of knee osteoarthritis.2.Observe the effect of Qianggubao decoction:24 SPF grade female SD rats,6 month aged,were randomly divided into Normal Group(NOR),Model Group(MOG),Observation Group(OBG),Positive Control Group(PCG),6 in each group.The rats in NOR were raised in routine way,MOG given double evaporate water,OBG was given Qianggubao decoction of 1 g/ml,PCG was given Jingutongxiaowan decoction of 0.5 g/ml. The dosage was given 0.5 ml/100g,according to the weight to calculate the dosage,twice a day, for 4 W.Results1.The establishment of model:The content of E2 in serum,NOR versus RUN and OVX versus MOG both decreased(P<0.05),proving the ovaries model successful.After 6 W,compared with NOR and RUN the content of CTX-Ⅱ in urine and cartilage Mankin score, OVX and MOG both higher (P<0.05,P<0.01),and compared to OVX,MOG was higher (P<0.05).The Mankin rating of MOG has reached the degree of fibrosis of articular cartilage and moderate damage,proving the model of primary osteoarthritis of the knee successful.2.Qianggubao Mechanism:After administration of 4 W the content of CTX-Ⅱ in urine and cartilage Mankin score,compared with NOR MOG,OBG and PCG were increased (P<0.01,P<0.05);compared with the MOG,OBG and PCG were lower(P<0.05);OBG and PCG no statistical difference.The content of E2 in serum,compared with NOR,MOG,OBG and PCG were lower(P<0.01);compared with the MOG,OBG increased(P<0.05);compared with OBG, PCG lower(P<0.05).Synovial fluid IL-1β,TNF-a levels and degree of synovial inflammation, compared with NOR MOG,OBG and PCG were higher(P<0.01,P<0.05);compared with the MOQOBG and PCG were lower (P<0.05),no statistical significance between OBG versus PCG.Conclusion1.The knee OA model is successfully established in rats by the method of bilateral ovariectomy combined with treadmill Running Exercise.2.Qianggubao decoction can relieve primary osteoarthritis knee joint cartilage matrix especially Ⅱ type collagen degradation process and improve the role of chondrocytes apoptosis.3.Qianggubao decoction promote the compensatory regeneration of estrogen, inhibit the synthesis of inflammatory cytokines IL-1β,TNF-a;so as to provide a good environment for the repair of articular cartilage injury,and play its role.
