Objective:Investigate the single nucleotide polymorphism loci rs6841581A.G for zhuang people in Guangxi, China, and discuss its relationship with intracranial aneurysmsMethods We studied 87 IA patients with saccular intracranial aneurysms and 111 unaffected controls. All research sbjects are Zhuang Living in Guangxi,China.Genomic DNA was isolated from peripheral venous blood.The rs6841581A.G polymorphism was assessed by nested PCR-restriction fragment length polymorphism and agarosegel electrophoresis method,and then validated through DNA sequencing.Results In the recessive model,the frequency of homozygous GG genotype in research group is significantly higher than in the control group (p= 0.020). Gender stratification analysis shows:distribution of genotypes and alleles frequency was no significant difference between female aneurysm group and control group.Frequency of allele G in the male aneurysm group is higher than the control group (P=0.040). In the recessive model,homozygous GG genotype had a significantly higher frequency in both single and multiple IA group than in the control group (p<0.05). Compare the single and multiple IA subgroups each other, genotypes and alleles frequency distribution between the two subgroups had no significant difference.Conclusions SNP rs6841581 is associated with the intracranial aneurysms of Zhuang in Guangxi,and GG genotype may be the susceptible genotype for IA.The male IA patients and patients with single or multiple IA are all related to the SNP rs6841581 polymorphism. The relationship between female IA patients and SNP rs6841581polymorphism need further research.
