| Irritable bowel syndrome is a form of abdominal pain or abdominal discomfort associated with changes in bowel habits characterized by functional bowel disease.It was lacked a clear etiology and pathogenesis of the disease. Epidemiological studies have shown that it was associated with disorders of intestinal flora.Objective:Based on the technique of 16S rDNA, we studied the microbial community structure of intestinal bacteria in the normal subjects and patients with IBS. To construct gut flora bacterial spectrum in different age and different traditional Chinese medicine Constitution of both normal subjects and patients with IBS, revealing the core bacterial spectrum of the two groups and the differences between them.Methods:This study collected stool samples of 8 healthy subjects,14 patients with IBS as the object of study in Yangzhou region. We choose high-throughput sequencing technology to detect bacterial diversity of 16S rDNA variable region of V1-V3.Results:(1)In the normal, the community sequence of participants in 31-50 years old had the highest diversity and abundance. The highest of the four types of bacteria in Intestinal flora are Firmicutes (59.300%), Bacteroidetes (32.900%), Proteobacteria (4.600%) and Actinobacteria (4.000%). The dominant bacteria of all age groups are Firmicutes and Bacteroidetes; however, there was no significant difference, In normals, we divided 21-60 into 21-50 and 51-60 groups, the constitution of Oscillospira are higher in 21-50 group than 51-60 group, and Bifidobacterium, Lachnospiraceae, Clostridium, Veillonella are lower in 21-50 group than 51-60 group. (2)In IBS group, the community sequence of 31-50years old had the highest diversity, but the abundance is the lowest. The community sequence of 20-30years old had the higest abundance, but the diversity is the lowest. The highest of the four types of bacteria in Intestinal flora were Firmicutes (65.300%), and Bacteroidetes (25.400%), Proteobacteria (5.400%) and Actinobacteria (2.200%). The dominant bacteria of all ages are Firmicutes and Bacteroidetes, there was no significant difference.(3)In the normal and IBS group, the common highest four types of bacteria in Intestinal flora were Firmicutes, Bacteroidetes, Actinobacteria, and Proteobacteria,and there was no significant difference.At the genus level, the constitution of Coprococcus, Veillonella, Streptococcus, Klebsiella, Escherichia, Paraprevotella are higher in normal groupthan IBS group, which had a statistically significant difference (P<0.050).(4) In normal, the constitution of the intestinal flora had no significant differences between normal constitution and Yang deficiency constitution at the phylum and genus levels. In IBS group, at the level of pylum, the constitution of Acidobacteria in Yang deficiency is lower than normal constitution, which had a statistically significant difference (p=0.018). At the level of genus, the constitution of Phascolarctobacterium in Yang deficiency is lower than normal constitution, which had a statistically significant difference (p=0.039).(5)In IBS group,the prevalence of B.adolescentis, B.longum, B.breve, and B.animalis were 78.600%ã€85.700%ã€71.400%ã€78.600% andthe prevalence were 71.400%〠85.700%ã€57.100%ã€71.400% in normal respectively (P>0.050). The prevalence of L.mucosaeã€L.salivariusã€L.reuteri in IBS were 35.700%ã€42.900%ã€21.400%, andthe prevalence were 28.600%ã€14.300%ã€0.000% in normal andthe difference was non-significant. There was low detection rate of Lactobacillus.Conclusions:(1)The intestinal flora of IBS group were different between normal group, the constitution of Coprococcus, Veillonella in normal are higher and Streptococcus, Klebsiella, Escherichia, Paraprevotella in normal are lower in IBS group than Normal group.(2)In normal, the structure of intestinal microflora were changed in the elderly, the constitution of Oscillospira are higher in 21-50 group than 51-60 group. Bifidobacterium, Lachnospiraceae, Clostridium, Veillonella are lower in 21-50 group than 51-60 group. However, the structure of intestinal flora havs not been altered in IBS by all age groups.(3)In IBS group, the constitution of Acidobacteria and Phascolarctobacterium were different between normal constitution and Yang deficiency constitution at the phylum and genus levels. |
